Acetic acid bacteria

Sixty-six acetic acid bacteria (AAB) were isolated from fourty-five flowers and fruits collected in Hochiminh City, Vietnam. Of the sixty-six, thirty-one isolates were selected as dihydroxyacetone (DHA)-producing AAB based on the reaction with Fehling’s solution and grouped into three groups by routine identification with phenotypic features.

Undifferentiated human promyelocytic leukemia HL-60 cells show little or no superoxide production, but generate a very low O2 – concentration upon incubation with all-trans-retinoic acid (ATRA). Its production reaches a maximum within 20 h, and thereafter is maintained at an almost con-stant level. The differentiated cells show phorbol 12-myri-state 13-acetate (PMA)-stimulatedNADPHoxidase activity consistent with the amount of gp91phox(phagocytic oxid-ase) expressed in the plasma membrane.

The ability of yeasts to grow in the presence of weak organic acid preservatives is an important cause of food spoilage. Many of the determinants of acetate resistance inSac-charomyces cerevisiae differ from the determinants of resistance to the more lipophilic sorbate and benzoate. Interestingly,we show in this study that hypersensitivity to both acetate and sorbate results when the cells have auxo-trophic requirements for aromatic amino acids.

Propionate is used to protect bread and animal feed from moulds. The mode of action of this short-chain fatty acid was studied usingAspergillus nidulansas a model organism. The filamentous fungus is able to grow slowly on propio-nate, which is oxidized to acetyl-CoA via propionyl-CoA, methylcitrate and pyruvate. Propionate inhibits growth of A. nidulanson glucose but not on acetate; the latter was shown to inhibit propionate oxidation.
