Certain strains of microalgae known as diatoms were shown, under appropriate conditions, to produce up to 60% of their cellular mass as lipids under certain growth conditions. Acetyl-Co-A carboxylase catalyses the first committed step in the synthesis of fatty acids, i.e., carboxylation of acetyl-CoA to malonyl-CoA. In present study we have isolated the two species of marine diatoms Odontella aurita and Chaetoceros muelleri from Vellar estuary Parangipettai Southest coast of India and studied the acetyl co-A carboxylase activity at different Si treatment nutrient medium.

Certain amino acids, like glutamine and leucine, induce an anabolic response in liver. They activate p70 ribosomal protein S6 kinase (p70S6K) and acetyl-CoA carboxylase (ACC) involved in protein and fatty acids synthesis, respectively. In contrast, the AMP-activated protein kinase (AMPK), which senses the energy state of the cell and becomes activated under metabolic stress, inactivates by phosphorylation key enzymes in biosynthetic pathways thereby conserving ATP.

Liver metabolism is influenced by hormones and nutrients. Amino acids such as glutamine or leucine induce an anabolic response, which resembles that of insulin in muscle and adipose tissue. In this work, the signalling pathways and the effects of insulin were compared to those of glutamine and leucine in isolated hepatocytes from normal and streptozotocin-diabetic rats.

Rat hearts were perfused for 1 h with 5 mMglucose with or without palmitate or oleate at concentrations characteristic of the fasting state. The inclusion of fatty acids resulted in increased activities of the a-1 or thea-2 isoforms of AMP-activated protein kinase (AMPK), increased phosphoryla-tion of acetyl-CoA carboxylase and a decrease in the tissue content of malonyl-CoA. Pyruvate carboxylase (PC) fromBacillus thermodenitrificans was engineered in such away that the polypeptide chainwas divided into two, between the biotin carboxylase (BC) and carboxyl transferase (CT) domains. The two proteins thus formed, PC-(BC) and PC-(CT+BCCP), retained their catalytic activity as assayed by biotin-dependent ATPase and oxamate-dependent oxalacetate decarboxylation, for the former and the latter, respectively.
