In the current study, the effect of USP7 on pluripotency maintenance of mouse ESCs (mESCs) has been investigated with the help of cell viability assay, morphological analysis, alkaline phosphatase (ALP) staining, qPCR analysis, and Western Blotting. 600 nM P5091 application, which showed no significant toxicity in mESCs, increased the total ubiquitinated protein amount as a proof of the accomplishment of proper USP7 inhibition.

The aim of this study was to examine the effect of UV irradiation on alkaline protease gene expression and induction of overproducing strains. We aimed also to determine the genetic variation among high protease productive mutants using RAPD-PCR. The promising strains in this study may be used in various economic industrial applications and for successive genetic improvement as well.

Alkaline keratinolytic protease producing Bacillus species was screened out from soil sample and characterized by studying morphological, cultural and biochemical properties. It was identified by using Burgey‟s manual of determinative bacteriology, identification flow charts as Bacillus alcalophilus. The isolate was employed for the production of alkaline protease by using production medium consisting of glucose and casein as carbon and nitrogen sources at pH 8.5 (Rao and Narasu, 2007).

Pseudomonas aeruginosa is considered as the epitome of an opportunistic pathogens of hospitalized patients. A total of 159 clinical specimens were recovered and molecular identified by PCR assay. The number of isolates which identified as P. aeruginosa was 101 clinical isolates. Considering the effect of 14 natural products/extracts on some virulence factors of MDR isolates, sub inhibitory concentration (½ MIC) of Syzygium aromaticum (clove) buds extract inhibit the activity of alkaline protease enzyme on skimmed milk agar.

Proteases are one of the most important groups of enzymes with wide range of industrial applications. The soil isolate Bacillus cereusAG1 produced alkaline protease under submerged cultivation condition. Maximum production of enzyme was achieved at pH 10.0 and temperature 30oC at 120 rpm. The supplementation of fructose as carbon source and combination of beef extract, yeast extract and peptone as nitrogen source improved enzyme production by 1.39 times.

One of the most important properties of the protease enzyme is to degrade the biofilm because removal of biofilms is very difficult. In industrial settings, both the inactivation and removal of biofilms are of huge concern. If only disinfection without the removal of attached biofilms occurs, the inactivated biofilm cells may provide an ideal environment for further adhesion and growth, resulting in a complex matrix. Microbial resistance to biocides and their negative environmental impact are the main reasons for finding alternative biofilm control strategies.

Acidic and alkaline proteases from visceral waste of Labeo rohita (Hamilton, 1822) were isolated, partially purified by ammonium sulphate precipitation followed by dialysis, their kinetics and characteristics studied. The purification fold increased from 1.24 to 2.49 and 1.19 to 1.55 in acidic and alkaline protease respectively along the purification steps.

Production of alkaline protease from B. licheniformis N-2 was studied in shake flask experiments in growth medium comprised of (g/l): glucose, 10; soybean meal, 10; K2HPO4, 5; MgSO4.7H2O, 0.5; NaCl, 0.5; CaCl2.2H2O, 0.5. Different process parameters, such as fermentation period, initial pH, incubation temperature, agitation speed, and size and age of inoculum, were optimized for the maximum yield of the alkaline protease.

In this study, using the Alcapred software, the metagenomic DNA sequences of the gut flora of Coptotermes gestroi were analyzed to identify the enzymes that were specifically adapted to alkaline condition. The results show that 737 of 943 ORFs (accounting for 72%) encoded proteases, 154 of 214 ORFs (holding 78%) encoded lipases and 338 of 575 ORFs (accounting for 59%) encoded cellulase and hemicelluase.

It is scientifically and industrially important 1 to clarify the stabilizing mechanism of proteases in extraordinary envi-ronments. We used subtilisins ALPI and Sendai as models to study the mechanism. Subtilisin ALPI is extremely sensitive to highly alkaline conditions, even though the enzyme is produced by alkalophilicBacillus, whereas sub-tilisin Sendai from alkalophilic Bacillusis stable under conditions of high alkalinity.

Achromobacterprotease I (API) has a unique region of aromatic ring stacking with Trp169–His210in close proxi-mity to the catalytic triad. This paper reveals the electrostatic role of aromatic stacking in the shift in optimum pH to the alkaline region, which is the highest pH range (8.5–10) among chymotrypsin-type serine proteases. The pH-activity profile of API showed a sigmoidal distribution that appears at pH 8–10, with a shoulder at pH 6–8.
