Antibiotic resistance genes

Staphylococcus aureus is a major problem in both the clinical setting and within the community. S. aureus can quickly develop resistance to a wide range of antibiotics through a number of different mechanisms, of which, using transporters located in the cell membrane to pump antibiotics out of the cell is the most serious concern.

The MinIONTM is a portable DNA sequencing device that can sequence 16S rRNA genes. 16S rRNA genes are found in all bacteria and can be used to identify bacterial species. By sequencing 16S rRNA genes and analyzing the sequences for antibiotic resistance genes, we can identify bacteria that are resistant to antibiotics.

Among the other 12 inpatient MRSA isolates which had the pvl gene, nine were sensitive to most of the non- beta lactam antibiotics, whereas remaining three were resistant to most of the antibiotics except vancomycin and linezolid. The presence of pvl gene can no longer be used to discriminate between CA-MRSA and HA-MRSA.

A novel cosmid (pABC6.5) whose DNA insert fromStrep-tomyces capreolus, the A201A antibiotic producer, overlaps the inserts of the previously reportedpCAR11 andpCAR13 cosmids, has been isolated. These two latter cosmids were known to contain the aminonucleoside antibiotic A201A resistance determinants ard2 and ard1, respectively. Together, these three cosmids have permitted the identifi-cation of a DNA stretch of 19 kbbetweenard1andard2, which should comprise a large region of a putative A201A biosynthetic (ata) gene cluster....

Horizontal gene transfer has been a major force for genome plasticity over evolutionary history, and is largely responsible for fitness-enhancing traits, including antibiotic resistance and virulence factors. In particular, for adap-tation of prokaryotes to extreme environments, lateral gene transfer seems to have played a crucial role.

The cationic peptide antibiotic polymyxin has recently been reevaluated in the treatment of severe infections caused by gram negative bacteria. Methods: In this study, the genetic determinants for capsular polysaccharide level and lipopolysaccharide modification involved in polymyxin B resistance of the opportunistic pathogen Klebsiella pneumoniae were characterized. The expressional control of the genes responsible for the resistance was assessed by a LacZ reporter system.
