Bacteria of silkworm

The present investigation was carried out at P. G. Department of Sericulture, Poonch campus, University of Jammu for isolation and identification of various diseases. Silkworm just like all other living organisms is susceptible to infection and diseases. Various pathogens including viruses (Nuclear Polyhydrosis, Cytoplasmic Polyhydrosis), bacteria (flacherie), fungi (muscardine) and protozoa (pebrine) cause various diseases to silkworm.

Among the silkworm diseases, white muscardine caused by Beauveria bassiana is the most common disease. The seven different chemicals viz., BKC (90%), pine Oil (100%), bleach (12%), sodium chlorite (80%), bleaching powder (30%), sanitech (2000ppm) and hydrated lime (80%) were tested and screened against fungus (beauveria bassiana and bacteria (bacillus and staphyllococcus) based on poison food technique with different concentrations such as 0.05%, 0.5%, 1%, 2%, 2.5%, 3%, 5%and 10% with three replications for each chemical concentration along with control.

The study revealed that, the aqueous, acetone and alcoholic extract of A.vasica at different concentrations (2, 4 and 6 %) were highly effective against three bacterial species.

Silkworm gut bacterial isolates were identified by using 16S rRNA gene sequence data analysis. Total viable count, spore count and spore percentage were tested on selective NA medium and recorded at different hours interval. The highest value of total viable count was recorded (420 x 108CFU/ml), spore count was higher (100 x 108CFU/ml) after 72 h of inoculation.

Azotobacter is an aerobic, free living bacteria fixes atmospheric nitrogen in soil besides producing plant growth promoting substance. Eight Azotobacter isolates were isolated and purified from different tasar rearing regions of West Singhbhoom, Jharkhand on Jenson media by using serial dilution technique. Molecular diversity among these isolates was analysed using RAPD primers.

Lactic acid bacteria are described as heterogeneous group of regular, gram-positive, rod shaped, non-motile, non-spore forming bacteria with absence of catalase enzyme. Laboratory study was undertaken to isolate and characterize the lactic acid bacteria from silkworm gut in phenotype basis. A total of 21 silkworm gut samples were collected from silkworm breeds, Double hybrid {(CSR 6 X CSR 26) X (CSR 2 X CSR 27)} obtained from Chawki rearing center (CRC) at Annur, Coimbatore.
