In this work, the nonlinear I-V characteristics associated with transport across the polysilicon emitter interface are extracted based on correction of the standard open-collector method. The nonohmic behavior of the interfacial resistance is evaluated to determine whether direct tunneling is the dominant transport mechanism. An approximate model for direct tunneling is proposed to achieve an accurate fit to the measured tunneling current traversing ultrathin oxides in the low bias regime, where an increased leakage current is commonly observed.

Epigenetic characterization of cell-free DNA (cfDNA) is an emerging approach for detecting and characterizing diseases such as cancer. We developed a strategy using nanopore-based single-molecule sequencing to measure cfDNA methylomes. This approach generated up to 200 million reads for a single cfDNA sample from cancer patients, an order of magnitude improvement over existing nanopore sequencing methods.

The application of brushwood fences along the Mekong deltaic coast has recently played a significant role in wave damping and promoting sedimentation. The results show a good agreement of wave heights and wave spectrum between the physical, vegetation and porosity models. Furthermore, the computational and physical model errors, such as BIAS and SI values, are less than 1 mm and 10%, respectively.

The thesis is structured as follows: Chapter 1 reviews the literature and outlines the importance of blood, clinical use of blood, current methods of blood collection including alternative methods and the benefits of DBS. Chapter 2 summaries the overall methods used in the project. Chapter 3 explains the utility of hemaPEN® from both the collection and analytical perspective. Chapter 4 expands on the HCT bias associated with standard DBS method. Chapter 5 expands on the use of DBS in other fields such as genomic and epigenomics.

The Phenomenon of codon usage bias exists in the genomes of prokaryotes and eukaryotes. The codon usage pattern is afected by environmental factors, base mutation, gene fow and gene expression level, among which natural selection and mutation pressure are the main factors. Distinguishing the driver mutations from somatic mutations in a tumor genome is one of the major challenges of cancer research. This challenge is more acute and far from solved for non-coding mutations. Here we present OncodriveFML, a method designed to analyze the pattern of somatic mutations across tumors in both coding and non-coding genomic regions to identify signals of positive selection, and therefore, their involvement in tumorigenesis.

Most research articles presenting new data analysis methods claim that “the new method performs better than existing methods,” but the veracity of such statements is questionable. Our manuscript discusses and illustrates consequences of the optimistic bias occurring during the evaluation of novel data analysis methods, that is, all biases resulting from, for example, selection of datasets or competing methods, better ability to fix bugs in a preferred method, and selective reporting of method variants.
