In this thesis, the author focussed on providing an application framework to be used in brain segmentation - the process of tumor detection. This constitutes setting up software bundled into a Window machine which is easily distributable. Moreover, a segmentation algorithm was implemented and tested using the provided software. In order to test the bundled software by implementing specific brain segmentation from the literature, the author used CT scans freely available from the database of 108 Central Military Hospital.

Wilson disease (WD), an inherited disorder affecting copper metabolism, is characterized by hepatic cirrhosis and neuronal degeneration, which result from toxic levels of copper that accumulate in the liver and brain, respectively. We reported previously that the 1.3-kb promoter of the WD gene contains four metal response elements (MREs). Among the four MREs, MREa plays the most important role in the transcriptional activation of the WD promoter.

d-Atracotoxins (d-ACTXs) from Australian funnel-web spiders differ structurally from scorpion a-toxins (ScaTx) but similarly slow sodium current inactivation and compete for their binding to sodium channels at receptor site-3. Characterization of the binding of 125 I-labelled d-ACTX-Hv1a to various sodium channels reveals a decrease in anity for depolarized (0 mV;Kd6.5 1.4 nM) vs.polar-ized ()55 mV;Kd0.60.2nM) rat brain synaptosomes.

We have recently identified an Ets element controlling over 90%of the basal expression of the human presenilin 1 (PS1) gene. We have also shown that Ets1 and Ets2 act as trans-activators of the PS1 gene by cotransfection experiments in SK-N-SH neuronal cells. The PS1 gene is widely but dif-ferentially expressed across tissues and the expression in brain appears to be restricted to neurons. This inactivation followed pseudofirst-order kinetics with a second-order rate constant of 168M )1 Æmin )1 .The spectrumofDABIA-labeled enzyme showed a characteristic peak of the DABIA alkyl-ated sulfhydryl group chromophore at 436 nm, which was absent from the spectrum of the native enzyme.

RasGRF is a family of guanine nucleotide exchange factors with dual spe-cificity for both Ras and Rac GTPases. In this study, using mouse brain extracts, we show that both RasGRF1 and RasGRF2 interact with micro-tubules in an in vitro microtubule assembly system and this binding is very tight. To characterize this association, recombinant purified proteins con-taining different regions of RasGRF1 were tested for their ability to bind microtubules preassembled from pure tubulin.

N-Glycosylation is a cotranslational and post-translational process of pro-teins that may influence protein folding, maturation, stability, trafficking, and consequently cell surface expression of functional channels. Here we have characterized two consensus N-glycosylation sequences of a voltage-gated K + channel (Kv3.1). Glycosylation of Kv3.1 protein from rat brain and infected Sf9 cells was demonstrated by an electrophoretic mobility shift assay.
