Circulating tumor cell (CTC) clusters play a critical role in carcinoma metastasis. However, the rarity of CTC clusters and the limitations of capture techniques have retarded the research progress. In vitro CTC clusters model can help to further understand the biological properties of CTC clusters and their clinical significance.

The present study reported the isolation, elucidation of compounds from the whole plant and their cytotoxic activities on Hep G2 (HB - 8065TM, hepatocellular carcinoma), A549 (CCL-185™, pulmonary adenocarcinoma), and MCF-7 (HTB - 22TM, breast cancer) cell lines. As a result, velutin exhibited the most significant cytotoxicity against Hep G2, A549, and MCF-7 cell lines with IC50 values of 34.00, 30.85, and 41.33 μg/mL, respectively.

We report here that earthworm can be used as an in vivo system to prescreen antimitotic compounds. The known antimitotic compound colchicine and the aqueous extract of Acorus calamus L. rhizome were used to examine this system. The antimitotic activity of colchicine and the rhizome extract was confirmed by methods with Allium cepa root tip, earthworm regeneration, and MTT (3-(4,5-dimethyl thiazolyl-2-yl)-2,5-diphenyltetrazolium bromide). An earthworm regeneration assay examined the regeneration ability of tissues from amputated regions of the earthworm.

Currently it is unclear how in situ breast cancer progresses to invasive disease; therefore, a better understanding of the events that occur during the transition to invasive carcinoma is warranted. Here we have conducted a detailed molecular and cellular characterization of two, patient-derived, ductal carcinoma in situ (DCIS) cell lines, ETCC-006 and ETCC-010.

Meinox’s small molecule library was also tested in vitro for pathologically distinct forms of cancer, including pancreatic adenocarcinoma PANC1, breast carcinoma MCF7, and lymphoblastic carcinoma RS4-11 cell lines. Analysis of this library at a dose of 1 μM allowed the discovery of potent, specific or broadly active anticancer compounds against pathologically distinct cancers. This study shows that in vitro analysis of different cancers or other phenotypic assays with Meinox small molecule library may generate novel and potent bioassay-specific compounds.

One lignan (7′S,8′R,8S)-4,4′-dihydroxy-3,3′,5,5′-tetramethoxy-7′,9-epoxylignan-9′-ol-7-one (1) together with four neolignans (7α,8α)-dihydrodehydrodiconiferyl alcohol 9-O-β-D-glucopyranoside (2), (7α,8α)-dihydrodehydrodiconiferyl alcohol 9′-O-β-D-glucopyranoside (3), icariside E3 (4), and icariside E5 (5) were isolated from Pouzolzia sanguinea. Their chemical structures were elucidated by ESI-MS, NMR spectra, as well as in comparison with the data reported in literature.

Chemical investigation of the lichen Parmotrema praesorediosum (Nyl.) Hale led to isolate six phenolic compounds including praesorediosic (1), orcinol (2), orselinic acid (3), lecanorin (4), isolecanoric acid (5) and virensic acid (6). Among them, compound 1 appeared to be found for the first time in the nature. The structure of these compounds was elucidated by spectroscopic analyses of HRESIMS and NMR as well as the comparison of their NMR data with those in the literature.

Their structures were established by means of physical data (ESI MS, 1D and 2D NMR) and evaluated for the cytotoxic activities against liver hepatocellular carcinoma (HepG2), human lung cancer (NCI-H460), human epithelial carcinoma (HeLa) and human breast cancer (MCF-7) cell lines. This is the first report on phytochemical studies of this plant.

Mutations in NBN, the gene for Nijmegen Breakage Syndrome (NBS), are thought to predispose women to developing breast cancer, but a breast cancer cell line containing mutations in NBN has not yet been described. The p.R215W missense mutation occurs at sub-polymorphic frequencies in several populations.

Along with de novo resistance, continued exposure to trastuzumab, an anti-human epidermal growth factor receptor 2 (HER2/neu) antibody, can lead to acquired resistance. In this study, we characterize a new anti-HER2/neu antibody resistant and metastatic mouse breast carcinoma cell line, TUBO-P2J.

Accumulating evidences suggest that tumors are driven by a small population of cells, termed “cancer stem cells” (CSCs), which may be resistant to current therapeutic approaches. In breast carcinoma, the CSCs have been identified as a CD44+ /CD24− cell population.

We previously identified dermicidin (DCD), which encodes a growth and survival factor, as a gene amplified and overexpressed in a subset of breast tumors. Patients with DCD-positive breast cancer have worse prognostic features. We therefore searched for specific molecular signatures in DCD-positive breast carcinomas from patients and representative cell lines.

Elevated basal, ligand-independent, Wnt signaling in some canine breast cancer cells is not caused by classical mutations in APC, β-Catenin or GSK3β but, at least partially, by enhanced LEF1 expression. We examined the expression and function of EGFR/HER-regulated pathways on the ligand-independent Wnt signaling.

In breast cancer, p63 expression has been found predominantly in basal-A (epithelial-type) triplenegative breast carcinomas (TNBC). To investigate the functional role of p63 in basal-A TNBC, we created MDA-MB468 cell lines with inducible expression of the two major N-terminal p63 isoforms, TAp63α and ΔNp63α.

Breast cancer is one of the most common cancer and a leading cause of death in women. Up to date the most commonly used breast cancer cell lines are originating from Caucasians or Afro-Americans but rarely cells are being derived from other ethnic groups. Here we describe for the first time the establishment of a naturally transformed breast cancer cell line, KAIMRC1 from an Arab woman of age 62 suffering from stage IIB breast cancer (T2N1M0).

α-Mangostin (αMG) is a natural substance that exerts a wide range of antitumor effects. Recently, we described that free αMG was able to dissociate multicellular tumour spheroids (MCTSs) generated from breast carcinoma cells and to reduce their cellular viability and motility.

Their structures were characterized by the combination of HR-MS, one and two dimensional NMR spectroscopic analyses. In addition, ergosterol peroxide and kansenone showed good cytotoxicity against four cancer cell lines, KB (human epidermal carcinoma), MCF7 (human breast carcinoma), SK-LU-1 (human lung carcinoma) and Hep-G2 (hepatocellular carcinoma). This is the first report on the chemical constituents and cytotoxic steroids from Ganoderma sp. in Laos.

Eosinophil cationic protein (ECP), one of the major components of basic granules of eosinophils, is cytotoxic to tracheal epithelium. However, the extent of this eect on other cell types has not been evaluatedin vitro.Inthis study, we evaluated the eect of ECP on 13 mammalian cell lines. ECP inhibited the growth of several cell lines including those derived from carcinoma and leukemia in a dose-dependent manner.

Tumour necrosis factor-related apoptosis-inducing ligand (TRAIL) has attractedmuch attention because of its ability to kill tumour cells. In this study, we demonstrated that treatment of QGY-7703 cells with the combination of TRAIL and etoposide resulted in synergistic cytotoxic effects. In dissecting the mechanism underlying this syner-gistic effect, we found that treatment with etoposide alone resulted in the upregulation of Bax, while the level of trun-catedBid (tBid)wasunchanged.
