Present study was undertaken to evaluate the anti-apoptotic effect of LEHD-FMK (caspase inhibitor) on buffalo bull sperm before and after semen freezing. Tris egg yolk extender was treated with Z-LEHD-FMK (0, 2, 4, 6, 10 and 20 µM) and used for extension of six aliquots of semen from each ejaculate to final concentration of 80 million sperm/ml. Sperm samples were analyzed for sperm motility, viability, plasma membrane integrity, mitochondrial membrane potential and PLA (Phospholipase) activity at pre-freeze and post thaw stages.

The present study was undertaken to investigate the effect of temperature humidity index (THI) on various seminal attributes in buffalo bulls. Forty one ejaculates (41) from two buffalo bulls of 4 to 6 years weighing 500 to 750 kgs reared at semen production centre, Department of Veterinary Gynaecology and Obstetrics, Pantnagar India, were used. Semen was collected twice a week through AV method for 6 months. The temperature and relative humidity of respective days was taken from meteorological department located nearby.

The objective of this review is to summarize and compare the work on effects of cooling rates and critical temperature range on post thaw semen quality of different animals. It could be concluded from the present review that optimum freezing protocol for buffalo bull, sheep, pig and stallion is not standardized, so far. However, till to some extent success has been achieved in cattle bulls.

Present study was conducted to evaluate the anti-apoptotic effect of caspase inhibitor (ZDEVD-FMK) supplementation in buffalo bull semen. Z-DEVD-FMK was supplemented with Tris egg yolk extender @ 2, 4, 6, 10, 20 µM. Pre-freeze and post thaw samples were evaluated in terms of % individual motility, % viability, % HOS reactive sperms, status of mitochondrial membrane potential and status of sperm membrane phosphatidylserine.

Four Swamp buffalo bulls aged 5 to 8 years were used to study the fresh semen as well as biochemical characteristics. A total of 60 ejaculates comprising 15 from each bull evaluated for fresh semen characteristics and a total of 40 ejaculates comprising 10 from each bull were used for evaluation and biochemical characterization.

The study was conducted to evaluate the combined effect of herbs on pre-freeze and postthaw sperm motion traits in subfertile buffalo bulls. Three subfertile buffalo bulls which were maintained on the basal ration along with this, they were orally supplemented with Panax ginseng roots, Shilajit, Withania somnifera roots, Tribulus terrestris fruits, Turnera diffusa leaves; Ptychopetalum olacoides bark each @ 400 mg/100 kg body weight and Pausinystalia yohimbe bark @ 300 mg/100 kg body weight of bulls for 60 days of treatment phase.

Present study was conducted with the objectives to identify single nucleotide polymorphism in sperm associated antigen 11 B(SPAG11B) gene and to analyze association between identified polymorphism with conception rate in Murrah bulls in ICAR-National Dairy Research Institute (NDRI) herd, Karnal. A 373 base pair region covering partial intron 2, exon 3 and partial intron3 of bovine SPAG11 gene was amplified using genomic DNA extracted from eighty six Murrah bulls and genotyped using sequencing and polymerase chain reaction- restriction fragment length polymorphisms (PCR-RFLP) methods.

Semen cryopreservation is a widely used technique across the world for conservation and proliferation of genetically superior germplasm through artificial insemination. Artificial insemination (AI) is the most conclusive tool to increase genetic improvement, control of venereal diseases and improve herd performance and productivity. For achieving the mission, successful and effective cryopreservation of semen is essential.

There is a need to improve current practices in Asia with regard to selection of cattle for breeding purposes, for both dairy and beef production. For many years, most of the countries in the region have been importing cows, bulls, and semen, largely from the temperate regions of the world, and using them to ‘upgrade’ the genetics of their existing herds of indigenous cattle for producing ability. However, and based on current evaluation of production levels and the productivity of cattle and buffalo, some doubts exist regarding the need and wisdom to continue this practice.
