C2C12 myotubes

In this paper, antidiabetic activities of glucomanno-oligosaccharides (GMO) were in vitro and in vivo investigated. Treatment with 100 μg/ml and 50 μg/ml of GMO for 1 hour caused 1.47-fold and 1.81-fold phosphorylation of AMPK, respectively. Oral administration of GMO (6 g/kg-1 of body weight day-1 ) lowered blood glucose levels (p < 0.05) at 120 min as compared to control group. These results suggested that GMO exhibited anti-diabetic effects via activation of AMPK and could be useful for diabetes prevention.

It is well known that obesity is increasing as worldwide epidemic and it is often accompanied by increased metabolic disorders. In the present study, we differentiate C2C12 myocytes into mature myotubes. Then the cells were treated with free fatty acid (FFA) to mimic the obese microenvironment.

In the present study, we use LPS to treat C2C12 skeletal muscle cells and observe expression of several inflammatory makers. Our results show that expression of inflammatory cytokine IL6 mRNA is strongly induced in the LPS-treated C2C12 cells compared to the control cells. Surprisingly, expression of the upper controller of inflammation TLR2 mRNA in the LPS-treated C2C12 cells is similar as that in the control cells.

Insulin like growth factor-1 (IGF-1) is established as an anabolic factor that can induce skeletal muscle growth by activating the phosphoinosi-tide 3-kinase⁄Akt⁄mammalian target of rapamycin (mTOR) pathway. Although this signaling pathway has been the subject of much study, the molecular mechanisms linking IGF-1 binding to mTOR activation remain poorly defined in muscle.
