Casein kinase 2α

We have previously reported that a new intronless gene for casein kinase 2α (CK2α), CSNK2A3, is expressed in human cells. The promoter of the well-known CK2α, CSNK2A1, displays characteristics of a housekeeping gene, whereas CSNK2A3 has a characteristic of a regulated promoter with two TATA boxes and a CAAT box.

Casein kinase 2 (CK2) is involved in various cellular events such as proliferation, apoptosis, and the cell cycle. CK2 overexpression is associated with multiple human cancers and may therefore be a promising target for cancer therapy.

Diabetes mellitus (DM) is a chronic disease found worldwide. Notably, BKS.Cg- Dock7m +/+ Leprdb/JNarl mice are useful animal models for studying type 2 diabetes mellitus (T2DM). In this study, we investigated casein kinase 2 alpha 1 (CSNK2A1) gene and protein expression in the liver tissues of mice at different ages (4, 16, and 32 weeks) using real-time quantitative polymerase chain reactions, western blotting, immunohistochemistry, and enzyme-linked immunosorbent assay.

Calmodulin (CaM) is phosphorylated in vitro and in vivo by multiple protein-serine/threonine and protein-tyrosine kinases. Casein kinase II and myosin light-chain kinase are two of the well established protein-serine/threonine kinases implicated in this process. On the other hand, within the protein-tyrosine kinases involved in the phosphorylation of CaM are receptors with tyrosine kinase activity, such as the insulin receptor and the epidermal growth factor receptor, and nonreceptor protein-tyrosine kinases, such as several members of the Src family kinases, Janus kinase 2, and p38Syk....

An earlier described CK2btes gene of Drosophila melano-gasteris shown to encode amale germline speci®c isoformof regulatory bsubunit of casein kinase 2. Western-analysis using anti-CK2btes Ig revealed CK2btes protein in Drosophila testes extract. Expression of a CK2btes± b-galactosidase fusion protein driven by the CK2btes pro-moter was found in transgenic ¯ies at postmitotic stages of spermatogenesis.

A number of findings have suggested the involvement of protein phosphorylation in the regulation of the epithelial Na + channel (ENaC).Arecent studyhas demonstrated that the C tails of thebandcsubunits of ENaC are subject to phosphorylation by at least three protein kinases [Shi, H., Asher, C., Chigaev, A., Yung, Y., Reuveny, E., Seger, R. & Garty, H. (2002)J. Biol. Chem.277, 13539–13547]. One of them was identified as ERK which phosphorylates bT613 andcT623 and affects the channel interaction with Nedd4....

The ability of protein kinase C and casein kinase 2 substrate in neurons (PACSIN)⁄syndapin proteins to self-polymerize is crucial for the simulta-neous interactions with more than one Src homology 3 domain-binding partner or with lipid membranes.

Using co-immunoprecipitation combined with MS analysis, we identified the a¢ subunit of casein kinase 2 (CK2) as an interaction partner of the atypical Rio1 protein kinase in yeast. Co-purification of Rio1p with CK2 from Dcka1 or Dcka2 mutant extracts shows that Rio1p preferentially interacts with Cka2p in vitro.
