Virus neutralization test (VNT) is widely used for serological survey of classical swine fever (CSF) and efficacy evaluation of CSF vaccines. However, VNT is a time consuming procedure that requires cell culture and live virus manipulation. C-strain CSF vaccine is the most frequently used vaccine for CSF control and prevention.

Infectious Bursal Disease (IBD) is an economically important and contagious viral disease of poultry. An attempt was made for development of a synthetic peptide based diagnostic reagent. Cell culture adapted viral antigen was prepared by successful passage on CEF and then ultracentrifugation to get partially purified virus. Confirmation of the virus in was done by RT- PCR. The antigen was then used for the production of hyper immune sera in rabbits. Sera samples were also collected from known IBD vaccinated and non-vaccinated birds.

Phylogenetic relationship of the CSF cell culture vaccine virus has also been established with other known CSF reference vaccine viruses, lapinized as well as cell culture adapted strains.

Resazurin dye-based colorimetric assay is simple and cost-effective to evaluate viability and cytotoxicity. This dye also used for evaluating proliferative responses of PBMCs against mitogens and antigens especially to assess the CMI response against vaccines and pathogens. However, the assay needs to be optimized for different cell types and animal species. So far, resazurin based assay is not available for sheep and goats. In the present study, the assay was optimized for PBMCs of goat with two density gradients viz., Histopaque 1.077 and 1.083, and different culture conditions.

In India, Foot and Mouth Disease (FMD) research started in the then Imperial Bacteriological Laboratory at Mukteswar as early as in 1929. The work on the production of vaccines using cell culture technique commenced at Mukteswar in early sixties initially in goat kidney/calf kidney cell cultures and subsequently by 1971-72 with the availability of the BHK 21 cell line in monolayer cultures. With the increased demand for the FMD vaccines, the Bangalore campus of IVRI was established in 1972 with the objective of large scale production in fermenters.
