In this study, the fruiting bodies of the X. nigripes were collected to isolate, propagate in the liquid medium, and then cultured to induce the fruiting body formation in the rice medium. The results showed that the X. nigripes was successfully isolated on PDAY (Potato Dextrose Agar Yeast Extract) medium, and the mycelium and spore of the X. nigripes had unique characteristics of the species.

This project is focused on studying the biological effects of low-power RF radiation on the expression and function of mechanosensitive ion channel protein (Piezo-1). To evaluate the effects of long-term exposures, cells were exposed at the frequency of 1800 MHz and power of 17 dBm for a period of 2 and 4 hrs using the transverse electromagnetic (TEM) cell exposure system. To study the effects of short-term RF radiation (10 min), exposures emitted from the mobile phone headset at the frequency of 845 MHz and powers of -15 dBm and -5 dBm were applied to cell cultures.

Deproteinated cheese whey was used for biomass production of probiotic strain Lactobacillus helveticus MTCC 5463 in a biofermenter. Optimization of growth parameters such as temperature, pH and time of incubation as well as nutrient supplementation of cheese whey was carried out using response surface methodology (RSM). Cheese whey supplemented with 0.95% yeast extract and 0.95% proteose peptone, inoculated with 6% (v/v) active culture of L. helveticus MTCC 5463 and fermented for 24 h at optimized temperature of 40ºC and pH 6.25 yielded 3.25 g/L dry cell biomass and 14.

An optogenetic upgrade for the tet - OFF system fused the inhibitor to the blue light-responsive B-LID degron and optimized the performance of this construct with regard to the number of Med25VBD repeats. Preparation and characterisation of attenuated cold-adapted influenza A reassortants derived from the A/Leningrad/134/17/57 donor strain presents the assay was capable of detecting interference where the infectivity ratio for A/Len/17 and A/Beij/32 was 1:7. Many groups of proteins play important roles in the cell’s response to various stresses. The molecular chaperone GroEL of Escherichia coli represents one such highly conserved family of stress proteins. We have observed that isolated GroEL complexes from stationary cultures contain various polypeptides that can be released from the chaperonin by GroES and/or ATP, and identified two such polypeptides as the proteins GatY and UP12.

The main component of cerebral amyloid angiopathy (CAA) in Alzheimer’s disease is the amyloid-b protein (Ab), a 4-kDa polypeptide derived from the b-amyloid protein precursor (APP). The accumulation of Ab in the basement membrane has been implicated in the degeneration of adjacent vascular smooth muscle cells (VSMC). However, the mechanism of Ab toxicity is still unclear. In this study, we examined the effect of substrate-bound Ab on VSMC in culture. The use of substrate-bound proteins in cell culture mimics presentation of the proteins to cells as if bound to the basement membrane.

An oxidosqualene cyclase cDNA, LcIMS1, was isolated from cultured cells of Luffa cylindrica Roem. by heterologous hybridization with cDNA of Glycyrrhiza glabra b-amyrin synthase. Expression of LcIMS1 in yeast lacking endogenous oxidosqualene cyclase activity resulted in the accumulation of isomultiflorenol, a triterpene. This is consistent with LcIMS1 encoding isomultiflorenol synthase, an oxidosqualene cyclase involved in bryonolic acid biosynthesis in cultured Luffa cells.

Mice expressing a C-terminal fragment of the prion protein instead of wild-type prion protein die from massive neuronal degeneration within weeks of birth. The C-terminal region of PrPc (PrP121–231) expressed in these mice has an intrinsic neurotoxicity to cultured neurones. Unlike PrPSc, which is not neurotoxic to neurones lacking PrPc expression, PrP121–231 was more neurotoxic to PrPc-deficient cells. Human mutations E200K and F198S were found to enhance toxicity of PrP121–231 to PrP-knockout neurones and E200K enhanced toxicity to wild-type neurones.

An exo-1,3-b-glucanase has been isolated from cultural filtrate of Trichoderma viride AZ36. The N-terminal sequence of the purified enzyme (m ¼ 61 ^ 1 kDa) showed no significant homology to other known glucanases. The 1,3-b-glucanase displayed high activity against laminarins, curdlan, and 1,3-b-oligoglucosides, but acted slowly on 1,3-1,4-b-oligoglucosides. No significant activity was detected against high molecular mass 1,3-1,4-bglucans. The enzyme carried out hydrolysis with inversion of the anomeric configuration.

The octadecaneuropeptide (ODN; QATVGDVNTDRPG LLDLK) and its C-terminal octapeptide (OP; RPGLLDLK), which exert anxiogenic activity, have been previously shown to increase intracellular calcium concentration ([Ca21]i) in cultured rat astrocytes through activation of a metabotropic receptor positively coupled to phospholipase C. Strictosidine glucosidase (SG) is an enzyme that catalyses the second step in the biosynthesis of various classes of monoterpenoid indole alkaloids. Based on the comparison of cDNA sequences of SG from Catharanthus roseus and raucaffricine glucosidase (RG) from Rauvolfia serpentina, primers for RT-PCR were designed and the cDNA encoding SG was cloned from R. serpentina cell suspension cultures. The active enzyme was expressed in Escherichia coli and purified to homogeneity. Analysis of its deduced amino-acid sequence assigned the SG from R. serpentina to family 1 of glycosyl hydrolases.
