In this study, we tested the ability of a specific culture medium supplemented with a cytokine cocktail to increase the number of total MNCs and thereby enrich CD34+UCHSCs. In addition, we investigated the effects of the bioactive peptide SL-13R on the proliferation of CD34+UC-HSCs at different culture time points and peptide doses.

The primary objectives of this study were to: Investigate the fouling potential, and characterise the organic components of the membrane foulant layers, of clarified activated sludge and lagoon effluent obtained from the WTP; Investigate the fouling propensities of EOM and AOM derived from a blue-green algal culture as it passed through the growth cycle, ie., for which the cells had not been subject to physical disruption; Determine any difference in the fouling propensity of EOM and AOM and relate it to organic composition.

Graphene oxide (GO) has been attracting tremendous attention in the past few years in various biomedical applications including; tissue engineering, cell culture, biosensing, bioimaging and therapeutics delivery due to unique features it possesses. In this thesis, we demonstrate the preparation of crumpled GO functionalized with polyethyleneimine (PEI) (CGO-PEI) via surface acoustic waves (SAW) to develop a nanotherapeutic delivery platform with efficient cellular uptake and good biocompatibility.

This project is focused on studying the biological effects of low-power RF radiation on the expression and function of mechanosensitive ion channel protein (Piezo-1). To evaluate the effects of long-term exposures, cells were exposed at the frequency of 1800 MHz and power of 17 dBm for a period of 2 and 4 hrs using the transverse electromagnetic (TEM) cell exposure system. To study the effects of short-term RF radiation (10 min), exposures emitted from the mobile phone headset at the frequency of 845 MHz and powers of -15 dBm and -5 dBm were applied to cell cultures.

The results showed that the RPA reaction, without requiring complex thermal cycles, was well-performed in the optimal conditions of 39°C within only 25 minutes. The limit of detection was identified as 310 fg of L. monocytogenes genomic DNA, which was 1000-fold more sensitive than the conventional PCR. RPA also succeeded to directly detect L. monocytogenes cells at a concentration as low as 2.5 × 101 Colony Forming Unit (CFU)/mL in pure cultures. In addition, RPA could accurately detect L. monocytogenes at 2.5 × 102 CFU/mL in milk without sample extraction or processing.

This study was carried out to evaluate the effect of Cobalt chloride (CoCl2) on cardiac mitochondrial function in an in vitro model. In the study, H9C2 cardiomyocytes were cultured in a medium containing different concentrations of CoCl2. Cell viability, cardiolipin content, mitochondrial function, and mitochondrial oxidative stress were assessed by using Cell Counting Kit-8 and suitable fluorescence kits.

Deproteinated cheese whey was used for biomass production of probiotic strain Lactobacillus helveticus MTCC 5463 in a biofermenter. Optimization of growth parameters such as temperature, pH and time of incubation as well as nutrient supplementation of cheese whey was carried out using response surface methodology (RSM). The result showed that only 6% single cancer cells developed into tumorspheres during culture process. Then, flow cytometry and immunofluorescence analysis was carried out to evaluate the expression of CSC markers CD44 and ALDH in samples taken from tumorspheres which arose from gastric CSCs. The results showed that gastric CSCs formed tumorspheres in 3D culture condition with a high expression of 92% and 40,1% for markers CD44 and ALDH, respectively.

An optogenetic upgrade for the tet - OFF system fused the inhibitor to the blue light-responsive B-LID degron and optimized the performance of this construct with regard to the number of Med25VBD repeats. Preparation and characterisation of attenuated cold-adapted influenza A reassortants derived from the A/Leningrad/134/17/57 donor strain presents the assay was capable of detecting interference where the infectivity ratio for A/Len/17 and A/Beij/32 was 1:7. Progeny viruses were characterised genetically by amplification of defined regions within each of the six internal genes by PCR and identification of the products by restriction enzyme analysis.
