In this study, the fruiting bodies of the X. nigripes were collected to isolate, propagate in the liquid medium, and then cultured to induce the fruiting body formation in the rice medium. The results showed that the X. nigripes was successfully isolated on PDAY (Potato Dextrose Agar Yeast Extract) medium, and the mycelium and spore of the X. nigripes had unique characteristics of the species.

The insect ecdysteroid receptor consists of a heterodimer between EcR and the RXR-orthologue, USP. We addressed the question of whether this heterodimer, like all other RXR heterodimers, may be formed in the absence of ligand and whether ligand promotes dimerization. We found that C-terminal protein fragments that comprised the ligand binding, but not the DNA binding domain of EcR and USP and which were equipped with the activation or DNA binding region of GAL4, respectively, exhibit a weak ability to interact spontaneously with each other.

Cys341 of carboxypeptidase Y, which constitutes one side of the solvent-accessible surface of the S1 binding pocket, was replaced with Gly, Ser, Asp, Val, Phe or His by site-directed mutagenesis. Tripeptidyl-peptidase II (TPP II) is a large (Mr106 ) tripeptide-releasing enzyme with an active site of the subtil-isin-type. Compared with other subtilases, TPP II has a 200 amino-acid insertion between the catalytic Asp44 and His264 residues, and is active as anoligomeric complex. This study demonstrates that the insert is important for the formation of the active high-molecular mass complex.
