In the current study, the effect of USP7 on pluripotency maintenance of mouse ESCs (mESCs) has been investigated with the help of cell viability assay, morphological analysis, alkaline phosphatase (ALP) staining, qPCR analysis, and Western Blotting. 600 nM P5091 application, which showed no significant toxicity in mESCs, increased the total ubiquitinated protein amount as a proof of the accomplishment of proper USP7 inhibition.

To investigate the function of circSMAD2 in GBC, the level of circSMAD2 in GBC cells was detected by RT-qPCR. CCK-8 assay was performed to investigate the cell viability.

Here, it is aimed to investigate the differences in the expression levels of let-7a, miR10b, miR-21, miR-125b, miR-145, miR-155, miR-200c, miR-221, miR-222 and miR-335, which associated with gene and proteins in MCF-7 (parental) and MCF-7s (Mammosphere/stem cell-enriched population/CD44+/CD24-cells) cells treated with paclitaxel. MCF7s were obtained from parental MCF-7 cells.

The increasing methicillin resistant Staphylococcus aureus (MRSA) infections pose a serious threat. Accurate and rapid detection of methicillin resistance is important for ensuring the prompt start of antibiotherapy and control of MRSA in the hospitals. Molecular detection of mecA gene is the gold standard for identification of MRSA isolates. However, many laboratories don’t have the capacity for molecular techniques. Resazurin is a dye used as an oxidation reduction indicator in bacterial cell viability assays.

Objective of the research was to validate the potential of Syzygium aromaticum and Kaempheria galanga against biofilm formation and plasmid borne drug resistance exhibited by Pseudomonas spp. Antimicrobial property of S.aromaticum and K.galanga on P.aeruginosa culture was assessed using the agar well diffusion method. Crystal violet assay was employed to determine the percentage inhibition of P.aeruginosa biofilm by the extracts following generation of biofilm on multiwell plates.

A comparative study on the anticancer effects of anthocyanins derived from banana cv. Nendran flower bracts against human colon (HT-29) and cervical cancer (HeLa) cells were investigated. In vitro assays by MTT, AO/EtBr and MMP revealed that anthocyanins extract inhibited the growth of both HT-29 and HeLa cells. The percentage of cell viability decreased linearly with increment of anthocyanins. The IC50 values for anthocyanins were 41.18 µg/ml for colon cancer cells and 46.50 µg/ml for the cervical cancer cells by MTT assay. The MMP assay indicated IC50 values of 71.18 and 106.

Resazurin dye-based colorimetric assay is simple and cost-effective to evaluate viability and cytotoxicity. This dye also used for evaluating proliferative responses of PBMCs against mitogens and antigens especially to assess the CMI response against vaccines and pathogens. However, the assay needs to be optimized for different cell types and animal species. So far, resazurin based assay is not available for sheep and goats. In the present study, the assay was optimized for PBMCs of goat with two density gradients viz., Histopaque 1.077 and 1.083, and different culture conditions.

The transcription factor p63 plays a pivotal role in the development and maintenance of epithelial tissues, including the ocular surface. In an effort to gain insight into the pathogenesis of keratitis caused by HSV-1, we determined the expression patterns of the p63 and Bax proteins in the Staatens Seruminstitute Rabbit Cornea cell line (SIRC). Methods: SIRC cells were infected with HSV-1 at various multiplicities and maintained for different periods of time. Virus replication was measured by indirect immunofluorescence assay and Western blot analysis.
