The objective of this study was to investigate the production of microbial cellulase by using Aspergillus oryzae and its application in the treatment of coffee. The cellulase producing method used for microbe medium was in submerged fermentation with malt extract medium.

In the present study it was aimed to obtain novel strains of actinomycetes that have the ability to promote plant growth. For this, seven soil samples from the rhizosphere of Humulus lupulus (Pazaryeri, Bilecik) were used and potential isolates were obtained. 16S rRNA genes of 30 isolates were amplified by PCR and sequenced. Eighteen isolates were found to be closely related to Streptomycetes spp. and they were tested for their proteolytic activity, cellulase activity, phosphate solubility, IAA production, biofilm formation, and growth in nitrogen-limited medium.

Cellulosimicrobium sp. MP1 was isolated from termites gut, showing ability to biosynthesize cellulase including endoglucanase, exoglucanase, -glucosidase and xylanase and therefore is potential candidate for enzymatic saccharification of lignocellulosic material. Cellulolytic bacteria were previously isolated from the termite guts and efforts were taken to optimize parameters for cellulase production. Activities of diversity of enzymes which include, CMCase, avicelase and filterpaperase (FPase) were studied from crude cellulase extracts. CMCase activity was found optimum at different temperatures (35°C, 40°C and 45°C) at pH 7 and after 48 hrs of incubation time.

In this study we examined diversity and activities of some actinomycetes isolated from mangroves in Giao Thuy, Nam Dinh Province. The distribution of actinomycetes in soil is strongly affected by external factors. Extra-cellular enzyme producing ability of mangrove actinomycetes is very high, particularly the ability of producing cellulase degrading filter paper (122/128 strains), CMC (110/128 strains), protease (83/128 strains) and amylase (78/128 strains).

Plant growth promoting bacteria has become of great interest to promote the crop growth and protect from phytopathogens. Total of 100 bacterial isolates were obtained form 45 different samples of rhizosphere soil as well as sugarcane parts. After extensive screening, eight isolates found as potential for multiple PGP traits were further investigated for cellulase, chitinase, HCN production potential and antifungal activity. Isolates F271 and F373were found to produce chitinase.

Cellulose degrading fungi play an important role in recycling of cellulosic materials. Enzymes in the gut of three different stages (juvenile, pre-clitellate, clitellate) of the earthworm species Perionyx ceylanensis, reared in the vermibed substrates namely, teak leaf litter (TLL), paper mill sludge (PMS) and pressmud (PM) in combination with cowdung were analysed. Specific activity of two hydrolytic enzymes viz., cellulase and polygalactouranase was assayed in the shattering and non-shattering zones of pod wall and lignin content in pod wall of soybean varieties at four harvesting stages viz., at physiological maturity, 5 DAPM, 10 DAPM and 15 DAPM. The cellulase enzyme activities was lowest at non-shattering zone of pod wall of the susceptible varieties as compare to the resistant varieties but highest at shattering zone of pod wall of the susceptible varieties indicating its role in pod shattering.

Characterization of cellulolytic microorganisms associated with naturally decomposing waste material
Thirty four samples of naturally decomposing waste material were collected from rural and urban parts of Western Maharashtra from which 64 bacteria, 49 fungi and 63 actinomycetes strains were isolated. The isolates were screened for cellulolytic activity on Czapek’s mineral salt agar media amended with carboxymethyl cellulose. Seventeen bacteria, thirty fungi and fifty seven actinomycetes isolates showed zone clearance indicating cellulase enzyme activity.

The compost, landfill waste, paddy soil and kitchen waste were used for development of microbial inoculum for the enhancement of biogas production from cattle dung under anaerobic batch conditions. Six digesters were set up containing, biogas slurry, compost, paddy soil, landfill waste and kitchen waste along with cattle dung in different concentration. The dehydrogenase activity, cellulase activity, N, P, K content, volatile solids, total solids, total volatile fatty acids, C/N ratio and biogas production was studied in samples at different time intervals.
