The cellobiohydrolase Pc_Cel7D is the major cellulase produced by the white-rot fungusPhanerochaete chrysosporium, constituting 10% of the total secreted protein in liquid culture on cellulose. The enzyme is classified into family 7 of the glycoside hydrolases and, like other family members, catalyses cellulose hydrolysis with net retention of the anomeric carbon configuration.

The basidiomyceteCoprinopsis cinereaproduces the glycoside hydrolase family 6 enzyme CcCel6C at low and constitutive levels. CcCel6C exhibits unusual cellobiohydrolase activity; it hydrolyses carboxymethyl cellulose, which is a poor substrate for typical cellobiohydrolases. Here, we deter-mined the crystal structures of CcCel6C unbound and in complex with p-nitrophenyl b-d-cellotrioside and cellobiose.
