PIKfyve is an evolutionarily conserved lipid and protein kinase enzyme that has pleiotropic cellular functions. The aim of the present study was to investigate the effects of phosphatidylinositol-3-phosphate 5-kinase (PIKfyve) inhibitor, YM201636, on nonsmall cell lung cancer (NSCLC) cells growth, tumorigenicity, and claudin (CLDN) expressions. Three NSCLC cell lines (Calu-1, H1299 and HCC827) were used to compare the effects of YM201636. Cytotoxic effects of YM201636 were analysed using XTT assay.

Claudin-4 is a transmembrane protein expressed at high levels in the majority of epithelial ovarian tumors, irrespective of subtype, and has been associated with tumor cells that are both chemoresistant and highly mobile. The objective of this study was to determine the functional role that claudin-4 plays in apoptosis resistance and migration as well as the therapeutic utility of targeting claudin-4 activity with a small mimic peptide.

Bacterial toxins have evolved to an effective therapeutic option for cancer therapy. The Clostridium perfringens enterotoxin (CPE) is a pore-forming toxin with selective cytotoxicity. The transmembrane tight junction proteins claudin-3 and -4 are known high affinity CPE receptors. Their expression is highly upregulated in human cancers, including breast, ovarian and colon carcinoma.

Bài viết cho thấy biểu hiện và xác nhận sự biểu hiện của protein dung hợp CPE30-GFP được cảm ứng bằng IPTG trong tế bào chủ E. coli BL21(DE3) bằng phương pháp SDS PAGE và lai Western với kháng thể kháng 6xHis Tag, cho thấy khả năng biểu hiện vượt mức protein CPE30-GFP trong tế bào chất và chủ yếu ở dạng tan. Đồng thời, CPE30-GFP đã bước đầu được tinh sạch với độ tinh sạch khoảng 92,3%. Thử nghiệm tương tác in vitro trên chip bán dẫn silicon đo điện trường (SiNW FETs) cho thấy CPE30 GFP tương tác tốt với thụ thể Claudin-4 (R4).

Trong nghiên cứu này, CPE16-GFP được thu nhận để làm nguyên liệu nhằm thử nghiệm khả năng bám dính với t´e bào M. Plasmid tái tổ hợp pET22b- cpe16-gfp được tạo ra thông qua việc nối gene cpe16-gfp vào plasmid pET22b đã xử lý với cặp enzyme cắt hạn chế NdeI và XhoI. Plasmid tái tổ hợp được hóa biến nạp vào E. coli BL21 (DE3) và cảm ứng biểu hiện với 0,5 mM IPTG. Protein CPE16-GFP được kiểm tra và xác định bằng phương pháp SDS-PAGE và Western blot với kháng thể kháng 6xHis.
