Ebook "Basic confocal microscopy" builds on the successful first edition by keeping the same format and reflecting relevant changes and recent developments in this still-burgeoning field. This format is based on the Confocal Microscopy Workshop that has been taught by several of the authors for nearly 20 years and remains a popular workshop for gaining basic skills in confocal microscopy.

Elucidation of protein transport mechanism in ion exchanges is essential to model separation performance. In this work we simulate intraparticle adsorption profiles during batch adsorption assuming typical process conditions for pore, solid and parallel diffusion. Artificial confocal laser scanning microscopy images are created to identify apparent differences between the different transport mechanisms.

The immobilization method and atomic concentrations of Si 2p, Au 4f, S 2p, C 1s, N 1s, and O 1s of the resulting SA–biotin modified Si surface were determined by XPS. The CD spectrum and confocal microscopy imaging confirmed that step-by-step modification and bioconjugation can be monitored successfully. Such detailed and well-defined step-by-step characterization provides good information about the surface properties of biosensor platforms. In addition, the LSPR sensing ability of the Au Np based platforms was studied by using a model SA–biotin system.

After studying the "Lecture Biological physics" you should be able to study of biological systems and biological processes using physics-based methods or based on physical principles. Biophysics discovers how atoms are arranged to work in DNA and proteins. Biophysicists are discovering how proteins work, they use these protein structures for discovering how biological machines work, in health and also in diseases. Biophysicists study life at every level, from atoms and molecules to cells, organisms, and environments.

Fibered confocal fluorescence microscopy (FCFM) allows in vivo investigation of pulmonary microstructures. However, the bronchial epithelium can only be imaged using exogenous fluorophores. The objective of this study is to compare methylene blue (MB) and acriflavine genotoxicity and to assess FCFM performance for in vivo imaging of precancerous lesions.

EGFR mutations are routinely explored in lung adenocarcinoma by sequencing tumoral DNA. The aim of this study was to evaluate a fluorescent-labelled erlotinib based theranostic agent for the molecular imaging of mutated EGFR tumours in vitro and ex vivo using a mice xenograft model and fibred confocal fluorescence microscopy (FCFM).

Absorption and refraction induced signal attenuation can seriously hinder the extraction of quantitative information from confocal microscopic data. This signal attenuation can be estimated and corrected by algorithms that use physical image formation models. Especially in thick heterogeneous samples, current single view based models are unable to solve the underdetermined problem of estimating the attenuation-free intensities.

Neural stem cells are motile and proliferative cells that undergo mitosis, dividing to produce daughter cells and ultimately generating differentiated neurons and glia. Understanding the mechanisms controlling neural stem cell proliferation and differentiation will play a key role in the emerging fields of regenerative medicine and cancer therapeutics. Stem cell studies in vitro from 2-D image data are well established.

Analysis of single cells in their native environment is a powerful method to address key questions in developmental systems biology. Confocal microscopy imaging of intact tissues, followed by automatic image segmentation, provides a means to conduct cytometric studies while at the same time preserving crucial information about the spatial organization of the tissue and morphological features of the cells.

Phosphorylated histone H2AX, also known as γH2AX, forms μm-sized nuclear foci at the sites of DNA double-strand breaks (DSBs) induced by ionizing radiation and other agents. Due to their specificity and sensitivity, γH2AX immunoassays have become the gold standard for studying DSB induction and repair.

The applications of multiphoton microscopy for deep tissue imaging in basic and clinical research are ever increasing, supplementing confocal imaging of the surface layers of cells in tissue. However, imaging living tissue is made difficult by the light scattering properties of the tissue, and this is extraordinarily apparent in the mouse mammary gland which contains a stroma filled with fat cells surrounding the ductal epithelium.

Plant cell death plays important roles during plant-pathogen interactions. To study pathogen-induced cell death, there is a need for cytological tools that allow determining not only host cell viability, but also cellular events leading to cell death with visualization of pathogen development.

Developmental biology relies to a large extent on the observation and comparison of phenotypic traits through time using high resolution microscopes. In this context, transparent model organisms such as the zebrafish Danio rerio in which developing tissues and organs can be easily observed and imaged using fluorescent proteins have become very popular.

Fluorescence microscopy allows an extensive variety of analyzes in structural biology. Until the beginning of the seventeenth century living beings were limited to macroscopic organisms only, and there was no knowledge of the microscopic universe. Epi-fluorescence and Confocal microscopy appears in 1995 by Marvin Minsky with the aim of increasing the contrast and constructing three-dimensional images by the use of an aperture, allowing a great definition.

When studiedin vitro, mitochondrial permeability transition (MPT) is associated with an increase in mitochondrial per-meability to solutes up to 1500 Da in mass and a loss of electrical potential difference across the inner mitochondrial membrane (Dwmit). The MPT has been implicated as being important in cellular calcium homeostasis, autophagy and cell death via necrosis and apoptosis.
