Southern rice black-streaked dwarf virus (SRBSDV) causes severe epidemical disease on rice with the infected area up to millions of hectares in South China and North and Central of Vietnam. So far, there are no effective, cheap, quick, and practicable methods for diagnosing SRBSDV. The conventional RT-PCR technique is the most popular method for detecting SRBSDV with high accuracy.

Sequencing errors are key confounding factors for detecting low-frequency genetic variants that are important for cancer molecular diagnosis, treatment, and surveillance using deep next-generation sequencing (NGS). However, there is a lack of comprehensive understanding of errors introduced at various steps of a conventional NGS workflow, such as sample handling, library preparation, PCR enrichment, and sequencing. In this study, we use current NGS technology to systematically investigate these questions.

Ureaplasma diversum has numerous virulence factors that contribute to pathogenesis in cattle, including Lipid-associated membrane proteins (LAMPs). Therefore, the objectives of this study were to evaluate in silico important characteristics for immunobiological applications and for heterologous expression of 36 LAMPs of U. diversum (UdLAMPs) and, also, to verify by conventional PCR the distribution of these antigens in strains of Brazilian states (Bahia, Minas Gerais, São Paulo, and Mato Grosso do Sul).

Dermatophytosis in calves is a major public and veterinary health concern worldwide because of its zoonotic potential and associated economic losses in cattle farms. However, this condition has lacked adequate attention; thus, to develop effective control measures, we determined ringworm prevalence, risk factors, and the direct-sample nested PCR diagnostic indices compared with the conventional methods of dermatophytes identification.

Sepsis and other infectious complications are major causes of mortality and morbidity in patients after cardiac surgery. Whereas conventional blood culture (BC) suffers from low sensitivity as well as a reporting delay of approximately 48–72 h, real-time multiplex polymerase chain reaction (PCR) based technologies like “SeptiFast” (SF) might offer a fast and reliable alternative for detection of bloodstream infections (BSI).

Crustacean hyperglycaemic hormone (CHH) is a neuropeptide that was originally identified in the X-organ/sinus gland complex of the eyestalks (ESs) in crustaceans. Several CHH isoforms and spliced variants were later identified in other tissues, and their functions have still not been completely unveiled. In this study, the identification and characterisation of the conventional CHH prepropeptide from the ESs of the littoral crab, Carcinus aestuarii, via rapid amplification of cDNA ends was reported.

The results showed that the RPA reaction, without requiring complex thermal cycles, was well-performed in the optimal conditions of 39°C within only 25 minutes. The limit of detection was identified as 310 fg of L. monocytogenes genomic DNA, which was 1000-fold more sensitive than the conventional PCR. RPA also succeeded to directly detect L. monocytogenes cells at a concentration as low as 2.5 × 101 Colony Forming Unit (CFU)/mL in pure cultures. In addition, RPA could accurately detect L. monocytogenes at 2.5 × 102 CFU/mL in milk without sample extraction or processing.

Massively parallel reporter assays (MPRAs) enable high-throughput functional evaluation of various DNA regulatory elements and their mutant variants. The assays are based on construction of highly diverse plasmid libraries containing two variable fragments, a region of interest (a sequence under study; ROI) and a barcode (BC) used to uniquely tag each ROI, which are separated by a constant spacer sequence.

The study was carried out on 160 Pseudomonas aeruginosa isolates. Samples had been cultured, identified and tested for sensitivity. A phenotypic test (Double disc synergy test) was applied on the collected MDR isolates for detection of MBL enzyme production. Conventional PCR was performed for the detection of the resistance gene(blaVIM). 100 isolates were multi-drug resistant according to (Modified Kirby Bauer disc diffusion method), with sensitivity to some drugs specially Amikacin (20%). Phenotypic test for MBL resulted in (38%) positive isolates and (62%) were negative ones.

In the present investigation a total of 100 faecal samples/rectal swabs were collected from dogs suffering with gastrointestinal disorders and showing the symptoms of canine parvovirus, like bloody diarrhoea, vomiting and pyrexia. The samples were tested initially with SYBR green dye based real time PCR with different combinations of the cycles and the results obtained were further confirmed by conventional PCR. We could able to standardize the real time PCR which was more helpful in screening of the faecal samples further in large numbers.

In the past decades, Lactobacillus species have been widely used in the market because they are thought to have biological properties and beneficial health effects. Conventional microbiological methods are often time consuming and labor-intensive. Real-time polymerase chain reaction (PCR) method will allow to identify and quantify rapidly Lactobacilli probiotics in food. In this study, the thermal shock protein target region (hsp60) was used to identify Lactobacillus acidophilus species. The limit of detection, specificity, and accuracy were of 104 , 100 %, 100 %, respectively.

This study included the isolation of Klebsiella pneumoniae bacteria from different sources in the environment and identification by using API-20E system and a conventional Polymerase chain reaction assay which detects Klebsiella pneumoniae wbbZ gene that involved in expression of D-galacton. The results of our study confirmed the positive identification of the bacteria using API-20E analytical system and also the presence of wbbZ gene in two of Klebsiella pneumoniae tissue invasive strains using conventional PCR assay.

Resistance to carbapenems are because of various mechanisms like mutations in the outer membrane proteins like Opr D causing decreased permeability of the drugs, the carbapenem hydrolyzing enzymes–carbapenemases and the efflux mechanisms. Carbapenem resistance was studied in 140 non-repeat clinical isolates of Pseudomonas aeruginosa in a tertiary care hospital. Imipenem resistance was detected using Kirby Bauer’s standard disc diffusion method. MIC for Imipenem was detected in all resistant isolates using E-test strips.

Analysis of high microsatellite instability (MSI-H) phenotype in colorectal carcinoma (CRC) is important for evaluating prognosis and choosing a proper adjuvant therapy. Although the conventional MSI analysis methods such as polymerase chain reaction (PCR) fragment analysis and immunohistochemistry (IHC) show high specificity and sensitivity, there are substantial barriers to their use.
