  • Shoot apices of Physalis minima L. were cultured on fifteen full strength MS medium supplemented with various concentrations of cytokinins (BAP, KIN and TDZ). Tissue culture responses namely elongation of existing shoot and differentiation of multiple shoots were observed from the cultured explants. The establishment of cultured explants was not affected by the cytokinin concentration in the medium but overall shoot apices showed better establishment on medium supplemented with BAP than other cytokinins.

  • Wheat is the second most produced staple food crop in world. An efficient method for in vitro micro propagation and genetic transformation of wheat are crucial for both basic and applied research. The present study was initiated with a view to develop a regeneration system in 4 genotypes of wheat cultivars HD2888, HD2329, DBW148, and DBW88 as a prerequisite to transformation. The mature embryos were excised from seeds and cultured on MS medium supplemented with high and low concentrations of cytokinins and auxins respectively.

  • An in vitro protocol for plant regeneration through organogenesis was established for Picrorhiza kurroa Royle Ex. Benth (Scrophulairaceae). The plant is a principle source of glycoside that is Picroside-I, Picroside-II and Kutkoside. Callus was induced on Murashige and Skoog basal medium supplemented with cytokinin (Kn or BA) and auxin (2,4-D/IBA/NAA) using leaf, petiole and stem explants. Shoots were regenerated on MS media with BA (1.5 to 2.5 mgl-1 ) combined with auxin (IBA or NAA) in the range of 0.5 to 2.5 mgl-1 . Rooting was initiated with full or half media supplemented with 0.

  • Lathyrus sativus L. (grass pea) is an important grain legume crop grown all over the world. It is very recalcitrant and difficult to regenerate under in vitro conditions. The present study determined the effect of preconditioning of cytokinins [10 and 20 mg L–1 thidiazuron (TDZ) and 2-isopentenyl adenine (2iP)] and postconditioning of dissimilar levels of TDZ-indole-3-butyric acid (IBA) and 2iP-IBA on in vitro shoot multiplication using embryonic node explants.
