Design of pcr primers

Strictosidine glucosidase (SG) is an enzyme that catalyses the second step in the biosynthesis of various classes of monoterpenoid indole alkaloids. Based on the comparison of cDNA sequences of SG from Catharanthus roseus and raucaffricine glucosidase (RG) from Rauvolfia serpentina, primers for RT-PCR were designed and the cDNA encoding SG was cloned from R. serpentina cell suspension cultures. The active enzyme was expressed in Escherichia coli and purified to homogeneity. Analysis of its deduced amino-acid sequence assigned the SG from R. serpentina to family 1 of glycosyl hydrolases.

We describe a strategy for systematic amplification of chitin synthase genes (chs) in the filamentous ascomycetes plant-pathogenBotrytis cinereausing PCR with multiple degen-erate primers designed on specific and conserved sequence motifs. Eight distinctchsgeneswere isolated, namedBcchs I, II, IIIa, IIIb, IV, V, VIandVII. They probably constitute the entire chsmultigenic family of this fungus, as revealed by careful analysis of six euascomycetes genomes.

A novel, type 1 ribosome-inactivating protein designated charybdin was isolated from bulbs of Charybdis maritimaagg. The protein, consisting of a single polypeptide chain with a molecular mass of 29 kDa, inhibited trans-lation in rabbit reticulocytes with an IC50of 27.2 nm. Plant genomic DNA extracted from the bulb was amplified by PCR between primers based on the N-terminal and C-terminal sequence of the protein from dissolved crys-tals.

Protein disulfide isomerase family proteins are known to play important roles in the folding of nascent polypeptides and the formation of disulfide bonds in the endoplasmic reticulum. In this study, we cloned two similar protein disulfide isomerase family genes from soybean leaf (Glycine maxL. Merrill cv. Jack) mRNA by RT-PCR using forward and reverse primers designed from the expressed sequence tag clone sequences.
