A 2 year old male Jamunapari goat weighing 30 kilograms was presented with complaints of chronic diarrhea, progressive weight loss and poor response to therapy. Faecal sample was collected aseptically and subjected for the presence of Mycobacterium avium subspecies paratuberculosis (MAP) by Ziehl-Neelsen staining and polymerase chain reaction (PCR).

Paratuberculosis (Johne‟s disease) is a chronic disease affecting livestock especially ruminants. The disease is caused by Mycobacterium avium subspecies paratuberculosis (MAP) an acid fast bacilli. The disease is characterized by emaciation, progressive weight loss and chronic enteritis unresponsive to treatment. This paper describes a mini review on diagnosis of Johne's disease.

Culture of clinical samples is the confirmatory test for the diagnosis of paratuberculosis but, it is time consuming due to its long incubation period, further it requires a skilled personnel to monitor the growth. Use of molecular techniques such as PCR is advantageous over the long incubation period in culturing of samples; more than eight months of incubation is needed to declare a sample to be negative.

Mycobacterium avium subspecies paratuberculosis (MAP) is a Gram positive, acid-fast bacterium which causes Johne’s disease or Paratuberculosis in domestic livestock species. For screening of cows for MAP infection a total of 103 milk samples from individual cows were collected from organized (57) and unorganized (46) farms. Presence of anti-MAP antibodies in milk were screened by indirect enzyme linked immunosorbent assays (iELISA) test, 72 (69.90%) cow milk samples were found positives (66.07% from organized farms and 74.46% from unorganized farms). Positivity percentages were 62.

Johne’s disease (JD) is caused by Mycobacterium avium Subsp. Paratuberculosis (MAP) and the disease is characterized by chronic diarrhea, progressive weight loss and emaciation. The present study was carried out to find out the MAP infection status by postmortem and molecular diagnosis in sheep and goats suspected for JD.
