Distinguishing barcodes

The new species is clearly distinguished from its congener Weygoldtia davidovi (Fage, 1946) by having more articles in the tibia and tarsus of leg I. A fragment of the cytochrome c oxidase subunit I (COI) gene is also provided for the new species. The K2P genetic distance of the COI between the new species and W. davidovi is 20%, and phylogenetic analysis also supported the new species.

DNA barcoding is a short DNA fragment, is used to distinguish between species, especially morphologically similar species and processed specimens that are difficult to distinguish species. For plants, the regions of DNA used as barcodes in identification or taxonomy are usually sequences of the chloroplast genome and the nuclear genome, including the coding regions and the non-coding regions.

An important challenge in pre-processing data from droplet-based single-cell RNA sequencing protocols is distinguishing barcodes associated with real cells from those binding background reads. Existing methods test barcodes individually and consequently do not leverage the strong cell-to-cell correlation present in most datasets.

Atractylodes DC is the basic original plant of the widely used herbal medicines “Baizhu” and “Cangzhu” and an endemic genus in East Asia. Species within the genus have minor morphological differences, and the universal DNA barcodes cannot clearly distinguish the systemic relationship or identify the species of the genus.

Huyen-sam (Vietnamese name) which belongs to Scrophularia L. genus is a valuable herb. This medicinal plant is classified as Scrophularia ningpoensis Hemsl. Huyen-sam roots, which contain a large amount of bioactive compounds, have a similar morphology to its relatives. DNA barcodes promise to be a precise and reliable tool for distinguishing the processed Huyen-sam materials from their counterfeits. In this paper we report a second species of Tomophagus from Cat Tien National Park in southern Vietnam (Cat Tien is a designated UNESCO Biosphere Reserve; The MAB Programme 2007). The new species reported here is distinguished from T. colossus on the basis of combined evidence from morphology, cultural characteristics, and ITS rDNA barcodes.
