DNA aneuploidy

Hiện tượng lệch bội nhiễm sắc thể ở phôi là một trong những nguyên nhân chính gây thất bại làm tổ, sẩy thai liên tiếp, làm giảm hiệu quả điều trị thụ tinh trong ống nghiệm. Xét nghiệm di truyền phôi tiền làm tổ phát hiện phôi lệch bội (Preimplantation Genetic Testing for Aneuploidy) giúp sàng lọc, phát hiện các phôi mang bất thường về số lượng nhiễm sắc thể bằng cách thu nhận 5-10 tế bào lá nuôi phôi (Trophectoderm).

Aneuploidies are copy number variants that affect entire chromosomes. They are seen commonly in cancer, embryonic stem cells, human embryos, and in various trisomic diseases. Aneuploidies frequently affect only a subset of cells in a sample; this is known as “mosaic” aneuploidy. A cell that harbours an aneuploidy exhibits disrupted gene expression patterns which can alter its behaviour. However, detection of aneuploidies using conventional single-cell DNA-sequencing protocols is slow and expensive.

Genetic information is physically carried on large DNA strings that are organized into chromosomes. Each species is characterized by a chromosome set that carry the information necessary and sufficient for its development and survival. Eukaryotic organisms are mostly diploid, containing two sets of chromosomes with each pair carrying nearly identical genetic information. Occasionally, exceptions to this rule are found, such as haploid yeast (with only one set of chromosomes) or polyploid ferns and frogs (with multiple sets).
