RNA aptamers with anity for the Ras-binding domain (RBD) of Raf-1 were isolated from a degenerate pool by in vitroselection.These aptamers eciently inhibited theRas interaction with the Raf-1 RBD, and also inhibited Ras-induced Raf-1 activation in a cell-free system. The RNA aptamer with the most potent inhibitory e€ect speci®cally inhibited the RasáRaf-1 interaction and had no anity for the RBD of the RGL protein, a homolog of the Ral GDP dissociation stimulator.

The light chain binding domain of rat myosin 1d consists of two IQ-motifs, both of which bind the light chain calmodulin (CaM). To analyze the Myo1d ATPase activity as a function of the IQ-motifs and Ca2+ ⁄CaM binding, we expressed and affinity purified the Myo1d constructs Myo1d-head, Myo1d-IQ1, Myo1d-IQ1.2, Myo1d-IQ2 and Myo1dDLV-IQ2. IQ1 exhibited a high affinity for CaM both in the absence and presence of free Ca 2+ .

Cys34 in domain I of the three-domain serum protein albumin is the bind-ing site for a wide variety of biologically and clinically important small molecules, provides antioxidant activity, and constitutes the largest portion of free thiol in blood. Analysis of X-ray structures of albumin reveals that the loop containing Tyr84 occurs in multiple conformations. In structures where the loop is well defined, there appears to be an H-bond between the OH of Tyr84 and the sulfur of Cys34.

Poly(A) binding protein (PABP) binds mRNApoly(A) tails and affects mRNA stability and translation. We show here that there is little free PABP in NIH3T3 cells, with the vast majority complexed with RNA. We found that PABP in NIH3T3 cytoplasmic lysates and recombinant human PABP can bind toAU-richRNA with high affinity. Human PABP bound an AU-rich RNA withKdin the nMrange, which was only sixfold weaker than the affinity for oligo(A) RNA.

Cytosolic phospholipase A2 -a (cPLA2 -a) is a calcium-activated enzyme involved in agonist-induced arachidonic acid release. In endothelial cells, free arachidonic acid is predominantly converted into prostacyclin, a potent vaso-dilator and inhibitor of platelet activation.
