  • In this study the effectiveness of plant growth regulators on micropropagation, total phenol content and metabolic profiles of Melissa officinalis L., important medicinal herb was assessed. The stem segments derived from one-month old in vitro germinated seedlings were used for initial explants. Of the eight different nutrient media tested the most favorable for micropropagation were found to be MP2 (MS medium enriched with 1mg/L BAP and 0.1 mg/L IBA) and MP3 (MS supplemented with 1.5 mg/L BAP and 0.5 mg/L NAA), which yielded 3.25- 4.0 shoots per explant within 4 weeks of culture.

  • The experiment consists of six treatments of growth regulators and control i.e. NAA 500 ppm, NAA 1000 ppm, NAA 1500 ppm, IBA 1000 ppm, IBA 1500 ppm and IBA 2000 ppm, were replicated four times in a complete randomized design (CRD). Presented data indicate that maximum number of shoots per cutting, maximum number of leaves per shoot, maximum number of roots per cutting, maximum length of roots (cm) were recorded under IBA 2000 ppm followed by IBA 1500 ppm, were significantly superior to the other growth regulators and control.

  • An effective protocol for in vitro proliferation and multiplication of Gerbera cv. Partrizia was established during 2015-17. Among the two stages initially used for culture initiation, capitulum explants cultured at immature stage responded better than the mature stage. Best establishment of immature capitulum explants and more number of quality shoots was obtained on modified MS medium supplemented with 10 mg/l BAP and 1 mg/l IAA. Half strength MS medium containing NAA (0.5 mg/l) + IBA (0.5 mg/l) was best for root induction.

  • This study was initiated to observe the effect of growth regulators (IBA 500, 1000, 1500, NAA 500, 1000, 1500 ppm) and growing medium combinations (Sand, sand 85% + manure 15%, sand 70% + manure 30%, sand 70% + manure 15% + soil 15%, sand 85% + soil 15%) on the rooting of three rootstocks of rose (Rosa indica, Rosa banksiae and Rosa bourboniana). Main effect of growth regulators and genotype was significant on majority of rooting characteristics. IBA (1000 ppm) recorded minimum days to root initiation (23.33), maximum rooting (72.22%), root length (6.42 cm) and field survival (82.38%).

  • A method for micropropagation was developed for Dendrocalamus asper, an economically and environmentally important bamboo. Disinfected seeds were cultured in flasks containing 20 ml of Murashige and Skoog’s medium (MS) supplemented with BA (1.0-7.0 mg l-1 ) or kinetin (1.0-7.0 mg l-1 ). Multiple shoots (6.53) were formed on MS medium supplemented with 3.0 mg l-1 BA and reached 1.49 cm in length. Continuous shoot proliferation was achieved on a MS medium supplemented with BA (1.0-7.0 mg l -1 ). The multiplication rate of 3.30 fold was achieved on MS medium supplemented with 3.0 mg l-1 BA.

  • The present study develops a protocol for rapid in vitro micropropagation of a critically endangered and floriculturally most important epiphytic orchid, Dendrobium primulinum Lindl. through the culture of small shoot tip explants (0.3 to 0.5mm) derived from in vitro grown seedlings. The shoot tip explants cultured on solidified Murashige and Skoog (MS) basal medium and MS medium alone or supplemented with combination of various concentrations of growth regulators; α-naphthalene acetic acid (NAA) and 6 benzylaminopurine (BAP), produced shoots and multiple shoots.

  • The present investigation was conducted to study the effects of auxin and cytokinin on callus induction from leaf blade, nodal, and runner segments of 3 strawberry clones. Among the 32 media combinations tested, 4.0 mg/L NAA + 1.5 mg/L IBA yielded the highest percentage of callus in all types of explants.
