This paper describes the development of electrochemical label- free DNA sensor for detection of transgenic in Round–up Ready Soybean (herbicide tolerance). Two pairs of electrodes which the optimized dimension is 70µm x 30µm (the width of electrode x inter-distance between fingers), were fabricated on a chip for differential measures.

The electrooxidation of 5-hydroxy-1,4-naphthoquinone (JUG) in organic media leads to conducting polymer film on glassy and Pt electrodes. The IR ex situ, IR in situ (MIRFTIRS) during the film formation, XPS studies indicate that electropolymerization occurs via the OH group while quinone group on the other ring remains intact and shows a very good electro activity and stability in different media, particularly in aqueous, biocompatible one.

For the first time, the detection of DNA sequences, related to HIV (found in AIDS patients), using Methylene Blue (MB) as the redox indicator was performed. The voltametric signals have been investigated on bared chitosan (CS) modified; probe immobilized (DNA single strand) and target-hybridized (DNA double strand) electrodes sequentially by means of Cyclic Voltammetry (CV) and Square Wave Voltammetry (SWV). The response of hybridization between the probe and perfectly complementary, double-base mismatch, single-base mismatch of analyzed targets were clearly identified.
