To establish an efficient in vitro regeneration protocol for orchardgrass (Dactylis glomerata L.), type of medium, several plant growth regulators and their various concentrations were evaluated in a factorial experiment for callus induction and plant regeneration. Endosperm supported mature embryo were used as explants and optimum response in callus induction was observed on Chu medium with 12 mg/L dicamba (3.6-dichloro-2-methoxybenzoic acid). The highest responding embryogenic callus (15%) was found with Murashige and Skoog medium and 8 mg/L 2.4-D (2,4-dichlorophenoxy acetic acid).

Anther culture is an effective breeding method for improving desirable traits, since it allows fast achievement of genetically homozygous lines. The present studies were carried out to evaluate the response of flue-cured tobacco anthers for high frequency pollen embryo induction in different culture media, and the effects of colchicine treatment on tobacco chromosome doubling. The results showed that different media caused significant changes in pollen embryo induction, in which the most obvious change was found in B medium with induction rates of 23.28%, 26.64%, and 28.

The seed containing an embryo, a miniature of new plant, structurally and physiologically equipped to sustain the growing seedling until it establish as new plant. Some traits are acquired during evolution for capacity to survive under unfavourable condition. One of such trait is ‘dormancy’, an intrinsic/extrinsic block to germination. The study was conducted to standardize an efficient protocol for callus induction, subsequent growth and in-vitro regeneration in six genotypes of proso millet viz., Gavhe, Sakhroli, Kalsuli, Vari No.10, Asond and Sukdhar. Wheat is the second most produced staple food crop in world. An efficient method for in vitro micro propagation and genetic transformation of wheat are crucial for both basic and applied research. The present study was initiated with a view to develop a regeneration system in 4 genotypes of wheat cultivars HD2888, HD2329, DBW148, and DBW88 as a prerequisite to transformation. The mature embryos were excised from seeds and cultured on MS medium supplemented with high and low concentrations of cytokinins and auxins respectively.

Somatic embryogenesis based plant regeneration is a pre-requisite for genetic transformation of banana. Mass multiplication of banana through somatic embryogenesis is also viewed as an economically viable alternative to commonly used shoot tip based method. Embryogenic cultures (EC) of Anatolian black pine (Pinus nigra J.F.Arnold subsp. nigra var. caramanica (Loudon) Businsky) were initiated from immature pre-cotyledonary zygotic embryos sampled weekly from 16 different trees in 2 consecutive years. Douglas-fir cotyledon revised (DCR) medium supplemented with 13.6 µM of 2,4-D and 2.2 µM of BAP was used for initiation and maintenance of EC.

Wheat embryogenic callus was induced by treating immature wheat embryos with 2,4-D. Digoxigenin-labeled germin sense and anti-sense RNA probes were prepared by in vitro transcription. By using these probes it was revealed that there was a striking induction of germin gene expression prior to visible callus formation.
