Embryo rescue is an in vitro technique that has been used to save the hybrid products of fertilization when they might otherwise degenerate. Embryo culture applied to practical problems is a tissue culture technique that has proven to be of greatest value to breeders. One of the main uses of this technique is to produce interspecific and intergeneric hybrids. Post-zygotic barriers such as endosperm abortion and embryo degeneration leading to low fertility have been overcome through the use of embryo culture and several hybrids have been developed in horticultural crops.

Successful interspecific hybrids were obtained when we crossed the cultivated soybean with wild annual Glycine soja. Although the success rate was low, whereas, the success rate was zero in case of wild perennial G. tomentella. Probably there is need to improve our crossability skills. Alternately, embryo rescue technique may be employed to recover the hybrids between cultivated soybean and wild perennial species. The hybridity between cultivated and wild annual soybean (Glycine soja) hybrids was confirmed at morphological and molecular level.

The present investigation entitled as “Effect of hardening media and different cross combinations on physiological parameters of in vitro cultured citrus plantlets” was carried out in the Division of Fruits and Horticultural Technology, ICAR-IARI, New Delhi. The experiment consisted of embryo rescued plantlets from cross pollination (control pollination) and open pollination (uncontrolled pollination) and two hardening media HM1(Coco peat: Perlite: Vermiculite, 1:1:1) and HM2 (Coco peat: Perlite: Vermiculite, 2:1:1).

Berseem, a nitrogen-fixing, annual, multicut forage crop cultivated around 2 million hectares areas of northern, central and eastern parts of India. Berseem has variability for pollination behavior however variation for morphological and agronomic traits are scare, perhaps, because of initially introduction of crop with narrow genetic base. Genetic variability have been fortified through introduction of exotic materials, intra-interspecific hybridization, induction of polyploidy and mutation. ICAR-Indian Grassland and Fodder Research Institute maintain >900 accessions of Trifolium spp.

In vivo induction of parthenogenesis using an irradiated pollen technique provides successful results in melon and accelerates breeding by shortening the time for obtaining 100% homozygous lines. Gamma rays (cobalt-60) are usually used for the pollen irradiation, although it is difficult to find a suitable source for irradiation.

This is the first report of successful regeneration of haploid lines in iris (Iris pseudacorus) developed by in situ parthenogenesis followed by embryo rescue. Native genotypes of I. pseudacorus (2n = 38) were prepared as female parents and I. spuria was set as the males.
