Ebook "Plant cell culture protocols (Third edition)" is taught around the world as part of undergraduate and graduate courses in agronomical and biological sciences. However, the apparent simplicity of cell culture technology should not lead to mistakingly consider it as trivial. It would be very diffi cult to understand modern plant biotechnology without in vitro cell cultures and even today, the application of recombinant DNA technology to important crops is hampered by the poor embryogenic or morphogenetic response of in vitro cultured tissues.

To establish an efficient in vitro regeneration protocol for orchardgrass (Dactylis glomerata L.), type of medium, several plant growth regulators and their various concentrations were evaluated in a factorial experiment for callus induction and plant regeneration. Endosperm supported mature embryo were used as explants and optimum response in callus induction was observed on Chu medium with 12 mg/L dicamba (3.6-dichloro-2-methoxybenzoic acid). The highest responding embryogenic callus (15%) was found with Murashige and Skoog medium and 8 mg/L 2.4-D (2,4-dichlorophenoxy acetic acid).

Genetic uniformity of in vitro raised plants is a prerequisite for production of quality planting material of banana. Embryogenic cultures were induced from immature male flower buds of two important cultivars of banana, ‘Rasthali’ (Silk-Musa AAB) and ‘Grand Naine’ (Cavendish-Musa AAA). Cell suspensions were established and regenerated plants were evaluated for their genetic stability using 10 inter simple sequence repeat markers.

Coconut (Cocos nucifera L.), is an important perennial crop cultivated across the globe. The demand for quality planting materials is increasing across the world for the establishment of new orchards, rejuvenation of existing orchards and production of quality nuts. Plant tissue culture is an alternate approach for the production of quality planting material. In this investigation, influence of auxin and cytokinin on somatic embryogenesis in coconut is studied using sliced mature embryos as explants in modified Y3 media supplemented with various concentrations of 2,4-D and kinetin.

Five maize lines prevalently used by breeders in the maize program were screened for their ability to form type II callus. Mature seeds were used as explant for in vitro culturing. One inbred line showed high embryogenic response and formed friable, type II calli and also exhibited successful plant regeneration. Ten culture media compositions based on Murashige and Skoog (1962) salts thus investigated differed with respect to the combinations of concentrations of auxins (IBA, NAA, picloram, 2, 4-D) and cytokinins (BAP, kinetin) were used.

Somatic embryogenesis based plant regeneration is a pre-requisite for genetic transformation of banana. Mass multiplication of banana through somatic embryogenesis is also viewed as an economically viable alternative to commonly used shoot tip based method. Banana somatic embryogenesis response was studied in three popular banana cultivars, Grand Naine, Rasthali and Hill banana „Virupakshi‟. Immature male flower bud explant was used for embryogenic callus induction.

Embryogenic cultures (EC) of Anatolian black pine (Pinus nigra J.F.Arnold subsp. nigra var. caramanica (Loudon) Businsky) were initiated from immature pre-cotyledonary zygotic embryos sampled weekly from 16 different trees in 2 consecutive years. Douglas-fir cotyledon revised (DCR) medium supplemented with 13.6 µM of 2,4-D and 2.2 µM of BAP was used for initiation and maintenance of EC.

Embryogenic mass was produced from kernels of mature fruits of Pistacia atlantica cultured in liquid Murashige and Skoog media, supplemented with 100 mg/l casein hydrolysate, 100 mg/l l-ascorbic acid, and benzylaminopurine (BAP). Embryogenic masses were differentiated directly from the kernel explants after culture for 3 weeks in liquid medium with 0.5-4 mg/l benzylaminopurine.

Salinity is one of the important abiotic stresses that restrict plant development. Triticale is a highly adaptable crop, known for its high-quality grain, yield potential, and resistance to disease.
