Enzymatic glutathione

The glyoxalase system usually comprises two enzymes, glyoxalase I (GLYI) and glyoxalase II (GLYII). This system converts cytotoxic methylglyoxal (MG) into non-toxic D-lactate in the presence of reduced glutathione (GSH) in two enzymatic steps. Recently, a novel type of glyoxalase III (GLYIII) activity has observed in Escherichia coli that can detoxify MG into D-lactate directly, in one step, without a cofactor.

The present study was planned to evaluate the effect of supplementing non-enzymatic antioxidants as adjunct therapy in enhancing clinical recovery of cattle naturally infected with bovine tropical theileriosis. A total of 18 crossbred dairy cattle with the clinical signs consistent of bovine tropical theileriosis and confirmation by detection of piroplasm infected erythrocytes in blood smears, were randomly divided into 3 groups (A, B, C) of 6 animals each.

Among the oysters, P. eous recorded highest levels of non-enzymatic antioxidative substances. Total glutathione content was significantly very high in P. eous, followed by P. sajor-caju, P. flabellatus and H. ulmarius. High level of total phenols, flavanoids, lycopene total carotenoid and vitamin A were recorded in P. eous. This was followed by P. sajor-caju, P. flabellatus and H. ulmarius. There was no significant difference in vitamin C content among the oyster mushrooms.

A study was conducted to investigate the effects of paclobutrazol (PBZ) (1.25 g a.i m-1 ) on the contents of non-enzymatic antioxidants, ascorbic acid, glutathione (GSH) and phenols and activities of antioxidant enzymes, peroxidase, catalase, superoxide dismutase and ascorbate peroxidase in buds and leaves of growing shoots of mango cv. Totapuri at four distinct phenological stages numerically represented as 510 (initiation of bud swelling), 511 (Swollen buds, 513 (bud burst) and 515 (panicle emergence) as per BBCH scale.

Antioxidant responses were analysed in Catharanthus roseus (L.) G.Don. under 0, 50 and 100 mM NaCl in order to investigate the plant’s protective mechanisms against long-term salt-induced oxidative stress. The NaCl treatments were repeated in 4 different stages of growth, i.e. 30, 45, 60 and 75 days after sowing (DAS).

This study investigated the enzymatic function of two putative plant GPXs, GPXle1 from Lycopersicon esculentum and GPXha2 from Helianthus annuus, which show sequence identities with the mammalian phospholipid hydroperoxide glutathione peroxidase (PHGPX). Both purified recombinant proteins expressed in Escherichia coli show PHGPX activity by reducing alkyl, fatty acid and phospholipid hydroperoxides but not hydrogen peroxide in the presence of glutathione.

Glutathione reductase (GR) recycles oxidized glutathione (GSSG) by converting it to the reduced form (GSH) using an NADPH as the electron source. The function of GR in the male genital tract of the rat was examined by meas-uring its enzymatic activity and examining the gene expression and localization of the protein.

The membrane associated proteins in eicosanoid and glutathione metabo-lism (MAPEG) superfamily includes structurally related membrane proteins with diverse functions of widespread origin. A total of 136 proteins belong-ing to the MAPEG superfamily were found in database and genome screenings. The members were found in prokaryotes and eukaryotes, but not in any archaeal organism.

The enzymatic activities of purified horseradish peroxidase, selenium-depen-dent glutathione peroxidase, thyroid peroxidase and myeloperoxidase, but not that of lactoperoxidase, were markedly enhanced when added into a reaction mixture containing 5lmnative seminal vesicle protein 4, a major protein secreted from rat seminal vesicle epithelium.
