Escherichia coli M15

The objective of this study is to successfully clone and express P46 of M. hyopneumoniae in Escherichia coli M15 cells to create an initial material source for developing effective solutions for the prevention and treatment of the PEP caused by M. hyopneumoniae bacteria.

The present study aims to investigate appropriate culture conditions for enhancing the expression of the cat1 gene encoding CAT1 enzyme from Burkholderia cepacia DF4 in Escherichia coli M15. The optimized culture conditions for gene expression are cell density at the time of induction, shaking speed, induction temperature, induction time, and inducer concentration. The highest level for CAT1 was obtained at the IPTG concentration of 1.2 mM, 10 hours after induction at 35 °C, shaking speed 200 rpm with cell density at OD600 0.7.

The rapid emergence of multiresistant microbial pathogens is a serious threat to human health. Extended spectrum beta lactamase (ESBL)-producing Escherichia coli is a superbug causing worldwide outbreaks, CTX-M has become the most commonly detected ESBL genotype; these enzymes have become predominant worldwide, the most common type of CTX-M reported in various geographic regions is CTX-M15. A total of 80 Escherichia coli (E. coli) isolates were recovered from different clinical samples of intensive care units (ICUs) patients.

Virus PED (PEDV, Porcine epidemic diarrhea virus) thuộc họ Coronaviridae là nguyên nhân gây dịch tiêu chảy cấp trên lợn. Bệnh lây lan rất nhanh và gây tỷ lệ chết cao; PEDV là một trong những mối lo ngại lớn của ngành chăn nuôi lợn trên toàn thế giới. Trình tự nucleotide mã hoá vùng quyết định kháng nguyên S1C và peptide CT24 được chứng minh có khả năng cảm ứng tạo kháng thể trung hòa, thích hợp cho việc phát triển vắc-xin PEDV tái tổ hợp. Gần đây, chủng PEDV mới (HUA-PED45) thuộc nhóm G2b được phát hiện tại Việt Nam gây thiệt hại lớn cho các trại chăn nuôi heo.
