Gelatin scaffold

Artificially fabricated hydroxyapatite (HAP) shows excellent biocompatibility with various kinds of cells and tissues which makes it an ideal candidate for a bone substitute material. In this study, hydroxyapatite nanoparticles have been prepared by using the wet chemical precipitation method using calcium nitrate tetra-hydrate [Ca(NO3)2.4H2O] and di-ammonium hydrogen phosphate [(NH4)2HPO4] as precursors. The composite scaffolds have been prepared by a freeze-drying method with hydroxyapatite, chitosan, and gelatin which form a 3D network of interconnected pores.

In present study, a previously fabricated tripolymer composite scaffold of PCL/gelatin/chitosan was modified with collagen I and the modified scaffold were studied for biocompatibility of L929 mouse fibroblasts and human osteoblast cells. Phase contrast microscopy confirmed the viability of L929 mouse fibroblasts and human osteoblasts cultured with the modified composite scaffold which indicated that scaffold did not produce any toxic effect on both cells.

Suitable three-dimensional scaffolds showing controllable drug delivery properties, known as multifunctional scaffolds, have gained increasing interest for soft tissue engineering. Here, tetracycline hydrochloride-encapsulated polylactic acid (TCH-PLA) microparticles prepared by using the double emulsion/solvent evaporation method were incorporated into a gelatin matrix.

. One approach in wound dressing development is to incorporate active molecules or drugs in the dressing. In order to reduce the frequency of dressing changes as well as to prolong wound healing efficacy, wound dressings that can sustain the release of the active molecules should be developed. In our previous work, we developed chitosan/sericin (CH/SS) microspheres that released sericin in a controlled rate. However, the difficulty of applying the microspheres that easily diffuse and quickly degrade onto the wound was its limitations.

In recent years, three-dimensional (3D) in vitro tumor models have become more common in cancer research across the world. Selecting scaffolds, seeding cells into the scaffolds, and culturing them are pivotal for the morphologies and proliferation of cancer cell lines. Therefore, this experience has developed a 3D tumor model based on co-culturing MCF-7 cells and fibroblasts (3:1) into gelatin-alginate scaffolds (3x3x5 mm3 ).

More than 200 polymers are used for electrospinning like silk fibroin (Zarkoob et al., 2004; Sukigara et al., 2003; Jin et al., 2004), collagen (Mathews et al., 2002), chitosan (Ohkawa et al., 2004), gelatin (Ma et al., 2005) etc. In the field of tissue engineering electrospinning technique is applied for the preparation of nanofiber scaffold design. The process is very versatile in terms of use of polymers, non-invasive and does not require the use of coagulation chemistry or high temperature for fiber generation.
