In this study, the Mgra16281 gene (ID: MK322955.1) enco ding an effector with the unknown function was cloned from the rice ro ot-knot nematode Meloidogyne graminicola isolated in Long An province. To knock-down the expression of this gene, an artificial microRNA was synthesized based on the Osa-MIR528 precursor and inserted into an expression vector.

The development of fruits without fertilization is known as parthenocarpy. Parthenocarpy improves the fruit quality, processing attributes, production and productivity of vegetable crops like tomato, cucumber, watermelon etc. Absence of seeds can enhances the shelf life of the fruits, allowing a better conservation, fruit set in adverse climatic conditions, early and offseason production of vegetable crops. Therefore, it is important to ensure yield stability regardless of environmental conditions.

The Sp family is a family of transcription factors that bind to cis-elements in the promoter regions of various genes. Regulation of transcription by Sp proteins is based on interactions between a GC-rich binding site (GGGCGG) in DNA and C-terminal zinc finger motifs in the proteins. In this study, we characterized the GC-rich promoter of the gene for the DNA methyltransferase (Dnmt1) that is responsible for methylation of cytosine residues in mammals and plays a role in gene silencing.

Synaptophysin, one of the major proteins on synaptic vesi-cles, is ubiquitously expressed throughout the brain. Syn-aptophysin and synapsin I, another synaptic vesicle protein, are also expressed by retinoic acid-induced neuronally dif-ferentiated P19 teratocarcinoma cells. Here, we show that inhibition of histone deacetylase activity in P19 cells is suf-ficient to activate transcription of the synaptophysin and synapsin I genes, indicating that neuronal differentiation and impairment of histone deacetylases results in a similar gene expression pattern....

Although ubiquitously expressed, the transcriptional factor CP2 also exhib-its some tissue- or stage-specific activation toward certain genes such as globin in red blood cells and interleukin-4 in T helper cells. Because this specificity may be achieved by interaction with other proteins, we screened a peptide display library and identified four consensus motifs in numerous CP2-binding peptides: HXPR, PHL, ASR and PXHXH. Protein-database searching revealed that RE-1 silencing factor (REST), Yin-Yang1 (YY1) and five other proteins have one or two of these CP2-binding motifs....

Lack of functional Fragile X mental retardation protein (FMRP) is the pri-mary cause of the Fragile-mental retardation syndrome in humans. In most cases, the disease results from transcriptional silencing of fragile mental retardation gene 1, fmr1, which encodes FMRP. However, a single mis-sense mutation (I304N) in the second KH domain of FMRP gives rise to a particularly severe case of Fragile X syndrome.

The human adenine nucleotide translocase-2 (ANT2) pro-moter contains a silencer region that confers partial repres-sion on the heterologous herpes simplex virus thymidine kinase (HSVtk) promoter [Barath, P., Albert-Fournier, B., Luciakova, K., Nelson, B.D. (1999)J. Biol. Chem.274, 3378–3384]. Two sequences in the silencer (Site-2 and Site-3) are protected in the DNase I assayin vitro, and one of these is a repeated GTCCTG element previously shown to act as theactive repressor element.

Báo cáo khoa học: Design of expression vectors for RNA interference based on miRNAs and RNA splicing
RNA interference (RNAi) mediates sequence-specific post-transcriptional gene silencing in many eukaryotes and is used for reverse genetic studies and therapeutics. RNAi is triggered by double-stranded small interfering RNAs (siRNAs), which can be processed from small hairpin RNAs gener-ated from an expression vector.

The nuclear envelope (NE) is composed of inner and outer nuclear mem-branes (INM and ONM, respectively), nuclear pore complexes and an underlying mesh like supportive structure – the lamina. It has long been known that heterochromatin clusters at the nuclear periphery adjacent to the nuclear lamina, hinting that proteins of the lamina may participate in regulation of gene expression.

RNA-mediated gene silencing (RNA interference) is a powerful way to knock down gene expression and has revolutionized the fields of cellular and molecular biology. Indeed, the transfection of cultured cells with small interfering RNAs (siRNAs) is currently considered to be the best and easi-est approach to loss-of-function experiments.

To elucidate the function of kappa class glutathione transferases (GSTs) in multicellular organisms, their expression and silencing were investigated in Caenorhabditis elegans. In contrast with most vertebrates, which possess only one GST kappagene, two distinct genes encoding GSTK-1 and GSTK-2 are present in theC. elegansgenome.

Báo cáo khoa học: In vivo RNA interference in oyster – vasa silencing inhibits germ cell development
This study investigated the potential of RNA interference, which is techni-cally challenging in bivalve mollusc species, to assess gene function in the oysterCrassostrea gigas. We designed dsRNA targeting the oyster vasa-like gene (Oyvlg), specifically expressed in oyster germ cells. In vivoinjection of oyvl-dsRNA into the gonad provokes a knockdown phenotype correspond-ing to germ cell underproliferation and prematurely arrested meiosis throu-gout the organ.

RNA silencing is an epigenetic inhibition of gene expression and is guided by small interfering RNAs. Sense transgene-induced post-transcriptional gene silencing (S-PTGS) occurs in a portion of a transgenic plant popula-tion. When a sense transgene encoding a tobacco endoplasmic reticulum x-3 fatty acid desaturase (NtFAD3) was introduced into tobacco plants, an S-PTGS line, S44, was obtained.
