The work presented in this thesis illustrates strategies to improve the detection of glucose by NanoZymes. This was done by first determining the NanoZyme activity of Cu and bimetallic nanoparticles of Cu, followed by the application of this activity towards biosensing of glucose.

Dietary sugars regulate expression of the intestinal Na + / glucose cotransporter, SGLT1, inmany species.Using sheep intestine as a model, we showed that lumenal monosaccha-rides, both metabolisable and nonmetabolisable, regulate SGLT1 expression. This regulation occurs not only at the level of transcription, but also at the post-transcriptional level. Introduction of D-glucose andsome D-glucose ana-logues into ruminant sheep intestine resultedin 50-fold enhancement of SGLT1 expression.

The addition of glucose toSaccharomyces cerevisiaecells causes reprogramming of gene expression. Glucose is sensed bymembrane receptorsaswell as (so far elusive) intracellular sensing mechanisms. The availability of four yeast strains that display different hexose uptake capacities allowed us to study glucose-induced effects at different glycolytic rates. Rapid glucose responses were observed in all strains able to take up glucose, consistent with intracellular sensing.

The AMP-activated protein kinase is an evolutionarily conserved hetero-trimer that is important for metabolic sensing in all eukaryotes. The muscle-specific isoform of the regulatory c-subunit of the kinase, AMP-activated protein kinasec3, has a key role in glucose and fat metabolism in skeletal muscle, as suggested by metabolic characterization of humans, pigs and mice harboring substitutions in the AMP-binding Bateman domains ofc3.
