In line with this, when we analyzed the amino acid sequence of E2F1, we found 3 hypothetical consensus PKA phosphorylation sites located at 127-130, 232-235, and 361-364 positions and RYET, RLLS, and RMGS sequences. After showing the binding and phosphorylation of E2F1 by PKA, we converted the codons of Threonine-130, Serine-235, and Serine-364 to Alanine and Glutamic acid codons on the eukaryotic E2F1 expression vector we had previously created.

The potassium inwardly rectifying channel, sub-family J, member 11 (KCNJ11), which is a pore-forming sub-unit of ATP sensitive potassium channels in pancreatic B-cells, plays a critical role in the regulation of insulin secretion. A non-synonymous E23K variant (SNP rs5219) in the KCNJ11 gene, which results from C to T transition and substitutes glutamate for lysine at position 23, is identified as a SNP associated with prediabetes and type 2 diabetes.

Huntington’s disease (HD) is an inherited neurodegenerative disorder characterized by cortico-striatal dysfunction and loss of glutamate uptake. At 7 weeks of age, R6/2 mice, which model an aggressive form of juvenile HD, show a glutamate-uptake deficit in striatum that can be reversed by treatment with ceftriaxone, a b-lactam antibiotic that increases GLT1 expression. Only at advanced ages ( 11 weeks), however, do R6/2 mice show an actual loss of striatal GLT1. Here, we tested whether ceftriaxone can reverse the decline in GLT1 expression that occurs in older R6/2s.
