Heterorhabditis bacteriophora

The efficacy of both botanical pesticides and entomopathogenic nematodes (EPN) is largely dose-dependent and driven by environmental conditions. Combination of the EPNs and botanicals may enhance their efficacy; thus, we investigated the compatibility of the medicinal plants Alepidea amatymbica and Elephantorrhiza elephantina with five locally isolated EPN strains, three Steinernema (S. khoisanae, S. biddulphi and S. innovationi) and two Heterorhabditis (H. bacteriophora and Heterorhabditis sp. SGI 244).

A total 200 soil samples were collected from tea plantation areas of district, Jorhat, Assam and were assessed for entomopathogenic nematodes using the Galleria baiting technique. Out of 200 soil samples, EPN was found in 1 soil sample collected from Tea plantation, Chetiagaon, Jorhat and designated as EPN-H-1. Morphological and morphometrical characters were used for the identification of nematode isolate. The measurements were expressed in % ratios and Means ± SD ranges.

Native isolates of EPNs, Heterorhabditis bacteriophora and Oscheius chongmingensis were isolated from the tea plantation crop of Jorhat district of Assam and were tested against tea mosquito bug Helopeltis theivora andbunch caterpillar, Andraca bipunctata through mortality test under laboratory condition. For that 5, 10, 50, 100,150, 200 and 250 infective juveniles of each isolate were inoculated per insect. The insect mortality was observed after 24, 48, 72 and 96h exposure.

The emergence in Galleria mellonella (L.) larva, which was infected by Steinernema feltiae and Heterorhabditis bacteriophora, began on the ninth day after exposure to the former nematode and the sixth day of exposure to the latter nematode (25 ± 0.5 ºC). The average number of nematodes developing in each host was as 13,829, and the range was from 4365 to 27,510 for S. feltiae infection.

The movement of 2 different isolates of an insect parasitic nematode species (Heterorhabditis bacteriophora Tur-H2 and H. bacteriophora Tur-H1) in vertical columns was tested under laboratory conditions. We have found significant differences in the movements of H. bacteriophora Tur-H2 and H. bacteriophora Tur-H1 towards the Galleria mellonella larvae as a host.

The effectiveness of Turkish strains of the entomopathogenic nematodes Steinernema feltiae and Heterorhabditis bacteriophora on the last instar larvae of Tenebrio molitor were investigated. Mortality rates of T. molitor larvae caused by each nematode species alone (experiment I) were determined.

Various application methods for the entomopathogenic nematode Heterorhabditis bacteriophora were evaluated under laboratory and field conditions. Four different methods of applying the infective juveniles (IJs) of the nematode to soil were assessed including insect cadavers (referred to as nematode-infected cadavers), subsurface injection, praying and drip irrigation.
