Ebook "Sphingosine-1-Phosphate: Methods and protocols (Second edition)" explores methods involving sphingosine-1-phosphate (S1P), a bioactive lysophospholipid which has become the focus of much research interest as it has widespread developmental and physio-pathological actions, controlling events within the nervous, reproductive, gastrointestinal, vascular, respiratory, and immune systems, in addition to having a prominent role in cancer, early mammalian embryogenesis, and stem cells.

For the first time a molecularly imprinted polymer (MIP)-based sensor for tyrosinase is described. This sensor is based on the electropolymerization of scopoletin or o-phenylenediamine in the presence of tyrosinase from mushrooms, which has a high homology to the human enzyme. The template was removed either by treatment with proteinase K or by alkaline treatment. The measuring signal was generated either by measuring the formation of a product by the target enzyme or by evaluation of the permeability of the redox marker ferricyanide.

A new series of acetamide derivatives containing 1,2,4-triazole and azinane moieties has been synthesized and characterized using 1H NMR, 13C NMR, IR, and EI-MS spectroscopic analysis. The intermediate triazole was synthesized through a sequential synthesis of carboxylate and carbohydrazide. The bovine serum albumin (BSA) binding of the newly synthesized 1,2,4-triazole derivatives was evaluated along with thermodynamics, site-selective binding, and synchronous study.

To evaluate the efficacy and safety of recombinant human serum albumin /granulocyte colonystimulating factor (rHSA/G-CSF) in breast cancer following receipt of cytotoxic agents. The prospective randomized controlled trials demonstrated that rHSA/G-CSF was efficacious and welltolerated with an approachable frequency and expense of application for prophylactic management of agranulocytosis.

Drug-protein binding may play a role in the thermal energetics of protein denaturation and could lead to the determination of its equilibrium dissociation parameter. The aim of this study was to assess the energetics of a drug that was bound to human serum albumin (HSA) during thermal denaturation. Drugs that were bound at a single high-affinity primary binding site on HSA, including diazepam and ibuprofen, were employed. Commercial HSA was treated with charcoal to remove stabilizers and adjusted to 20% w/v in a pH 7.4 buffered solution. Serial concentrations of individual drugs up to 0.

GSK’s varicella vaccine contains human serum albumin (HSA) which is used to stabilize the virus and prevent immunogens from adhering to the injection vial walls. However, because HSA is derived from human blood, there is a theoretical risk that it might contain infectious agents which could be unsafe for humans.

A previous study showed that the vaccine was immunologically non-inferior to the HSA-containing vaccine and well-tolerated in toddlers; low-grade fever was numerically higher in children receiving the vaccine without HSA, but the study lacked power to conclude on this difference.

Diabetes mellitus (DM) is one of the most common endocrinopathies observed in dogs. The main clinical manifestations are polydipsia, polyuria, polyphagia, weight loss and glucosuria. The etiology of diabetes mellitus is similar in dogs, cats and humans and is probably multifactorial (genetic, immune and environmental factors). Diabetes mellitus is described to be deficiency or absolute lack of insulin secretion is divided into two types: insulin dependent diabetes mellitus (IDDM) and non-insulin dependent diabetes mellitus (NIDDM). Various breed susceptibilities have been reported.

Chromium has been recognized as a new and important micro-nutrient, essential for both human and animal nutrition. This study was conducted to evaluate the appropriateness and/or the use of safety level of dietary chromium picolinate (Cr-Pic), and its effects on the physiological responses, the histometric characteristics, and the chemical analysis of dorsal muscles of mono-sex Nile tilapia, Oreochromis niloticus. niloticus. A total of 420 fingerlings (28.00 ± 0.96 g) were randomly distributed into 21 fiberglass tanks representing seven treatments at a rate of 20 fish m3 .

Inflammation and oxidative stress are two faces of one coin in end stage renal disease patients (ESRD) on maintenance hemodialysis. Their interconnection induces anemia complicated with erythropoietin hyporesponsiveness. The biochemical bases behind the resistance to erythropoietin therapy with frequent hemoglobinemia, oxidative stress and iron status have not been fully understood. Here two equal groups (40 patients each) of responders and non-responders to recombinant human erythropoietin therapy (higher than 300 IU/kg/wk of epoetin) were inves-tigated.

For the purpose of obtaining information about interphases between subunits of human serum albumin (HSA), a new tritium labelling method was adopted. The existence of linear dependence between accessible surface of amino acid residues and their specific radio-activity enabled to obtain information about qualitattive and quantitative amino acid composition of surface layer of protein globule and its large fragments.

A comparative study of thermodynamic and kinetic aspects of Cu(II) and Ni(II) binding at the N-terminal binding site of human and bovine serum albumins (HSA and BSA, respectively) and short peptide analogues was performed using potentiometry and spectroscopic techniques. It was found that while qualitative aspects of interaction (spectra and structures of complexes, order of reactions) could be reproduced, the quantitative parameters (stability and rate constants) could not.

Human serum albumin (HSA) is best known for its extraordinary ligand binding capacity. HSA has a high affinity for heme and is responsible for the transport of medium and long chain fatty acids. Here, we report myri-state binding to the N and B conformational states of Mn(III)heme–HSA (i.e. at pH 7.0 and 10.0, respectively) as investigated by optical absorbance and NMR spectroscopy. At pH 7.0, Mn(III)heme binds to HSA with lower affinity than Fe(III)heme, and displays a water molecule coordinated to the metal....

Cys34 in domain I of the three-domain serum protein albumin is the bind-ing site for a wide variety of biologically and clinically important small molecules, provides antioxidant activity, and constitutes the largest portion of free thiol in blood. Analysis of X-ray structures of albumin reveals that the loop containing Tyr84 occurs in multiple conformations. In structures where the loop is well defined, there appears to be an H-bond between the OH of Tyr84 and the sulfur of Cys34.

The changes in the far-UV CD signal, intrinsic tryptophan fluorescence and bilirubin absorbance showed that the guanidine hydrochloride (GdnHCl)-induced unfolding of a multidomain protein, human serum albumin (HSA), followed a two-state process. However, using environment sensitive Nile red fluorescence, the unfolding and folding pathways ofHSA were found to followa three-state process andanintermediatewas detectedinthe range0.25–1.5M GdnHCl.
