Hybrid molecules

Concept of hybrid molecules: When double strand DNA is steamed to a temperature exceed the melting temperature (Tm), it will separate into 2 single strands DNA due to breaks of H bonds. If the reaction temperature is then decreased slowly plus other appropriate experimental conditions, these ssDNA will pair again. This phenomenon is called the hybridization of molecules. Characteristic of the hybrid molecules: Specificity, the re-pairing occurs only between two complementary sequences.

In the further research and synthesis of hybrid molecules which exhibit highly biological activities, the synthesis of triazole-fused bicyclic heterocycles bearing the benzofuran has been undertaken. A facile, convenient and good yielding synthesis of novel benzofuran-bearing 1,2,4-trazole derivatives were described.

In the aim of further research of the highly bioactive benzofuran and triazine derivatives, the synthesis of 1,2,4-triazine derivatives bearing the benzofuran moiety has been undertaken. Using a simple and convenient method, a three steps synthesis of novel functionalized 1,2,4-triazine derivatives from readily available starting materials has been described.

The decoy approach against nuclear factor kB (NF-kB) is a useful tool to alter NF-kB dependent gene expression using synthetic oligonucleotides (ODNs) carrying NF-kB specific cis-elements. Unfortunately, ODNs are not stable and need to be extensively modified to be used in vivo or ex vivo. We have previously evaluated the possible use of peptide nucleic acids (PNAs) as decoy molecules. The backbone of PNAs is composed of N-(2-aminoethyl)glycine units, rendering these molecules resistant to both nucleases and proteases.

The siglecs (sialic acid-binding Ig-like lectins) are a distinct subset of the Ig superfamily with adhesion-molecule-like structure. We describe here a novel member of the siglec protein family that shares a similar structure including five Ig-like domains, a transmembrane domain, and a cytoplasmic tail containing two ITIM-signaling motifs. Siglec10 was identified through database mining of an asthmatic eosinophil EST library. Using the Stanford G3 radiation hybrid panel we were able to localize the genomic sequence of siglec-10 within the cluster of genes on chromosome 19q13.

RNA helicase A (RHA), a member of DNA and RNA helicase family containing ATPase activity, is involved in many steps of gene expression such as transcription and mRNA export. RHA has been reported to bind directly to the transcriptional coactivator, CREB-binding protein, and the tumor suppressor protein, BRCA1, and links them to RNAPolymerase II holoenzyme complex. Using yeast two-hybrid screening, we have identified RHA as an interacting molecule of the p65 subunit of nuclear factorjB(NF-jB).

The Protein Data Bank contains the description of approximately 27 000 protein–ligand binding sites. Most of the ligands at these sites are biologi-cally active small molecules, affecting the biological function of the protein. The classification of their binding sites may lead to relevant results in drug discovery and design.

The human lmo2 gene plays important roles in hematopoiesis and is associated with acute T lymphocyte leukemia. The gene encodes two protein isoforms, a longer form LMO2-L and a shorter form LMO2-S.
