  • The shoots in vitro were transferred to half-strength MS supplemented with different concentrations of IAA, IBA and NAA to investigate root formation. The best rooting occurred at 0,5 mg/L NAA (7.91 roots/shoot; root length of 4.01 cm; 98.51% root formation). The plantlets with uniform growth were planted on different substrate: Eco clean soil, Coconut fiber, Fern fiber, 50% Rice husk in combination with 50% Eco clean soil for research the most suitable substrate.

  • The present investigation was carried out at Research Farm, Division of Floriculture and Landscape Architecture, SKUAST-Kashmir, Shalimar, Srinagar, J&K (India) during the year 2010 to evaluate the effect of two growth regulators IBA and NAA @ 500, 1000, 1500 ppm in comparison to control (without any growth regulator treatment) and growing medium combinations (sand 100%, sand 85% + manure 15%, sand 70% + manure 30%, sand 70% + manure 15% + soil 15%, sand 85% + soil 15%) on the rooting of three rootstocks of rose (Rosa indica, Rosa banksiae and Rosa bourboniana).

  • The experiment consists of six treatments of growth regulators and control i.e. NAA 500 ppm, NAA 1000 ppm, NAA 1500 ppm, IBA 1000 ppm, IBA 1500 ppm and IBA 2000 ppm, were replicated four times in a complete randomized design (CRD). Presented data indicate that maximum number of shoots per cutting, maximum number of leaves per shoot, maximum number of roots per cutting, maximum length of roots (cm) were recorded under IBA 2000 ppm followed by IBA 1500 ppm, were significantly superior to the other growth regulators and control.

  • An effective protocol for in vitro proliferation and multiplication of Gerbera cv. Partrizia was established during 2015-17. Among the two stages initially used for culture initiation, capitulum explants cultured at immature stage responded better than the mature stage. Best establishment of immature capitulum explants and more number of quality shoots was obtained on modified MS medium supplemented with 10 mg/l BAP and 1 mg/l IAA. Half strength MS medium containing NAA (0.5 mg/l) + IBA (0.5 mg/l) was best for root induction.

  • Present investigation was carried out to determine the optimum level of different plant growth regulators for rapid shoot multiplication, rooting and hardening of micropropagated plantlets of Aloe vera. The experiment was laid out using completely randomized design (CRD) with ten replications during 2018-19. Auxiliary shoot explants were inoculated on MS medium supplemented with various concentrations of cytokinin (BAP 0.5 – 6.0 mg/l) alone and BAP (0.5-6.0 mg/l) with auxin (NAA 0.1 – 0.6) in combination for direct shoot bud proliferation.

  • This study was initiated to observe the effect of growth regulators (IBA 500, 1000, 1500, NAA 500, 1000, 1500 ppm) and growing medium combinations (Sand, sand 85% + manure 15%, sand 70% + manure 30%, sand 70% + manure 15% + soil 15%, sand 85% + soil 15%) on the rooting of three rootstocks of rose (Rosa indica, Rosa banksiae and Rosa bourboniana). Main effect of growth regulators and genotype was significant on majority of rooting characteristics. IBA (1000 ppm) recorded minimum days to root initiation (23.33), maximum rooting (72.22%), root length (6.42 cm) and field survival (82.38%).

  • An in vitro protocol for plant regeneration through organogenesis was established for Picrorhiza kurroa Royle Ex. Benth (Scrophulairaceae). The plant is a principle source of glycoside that is Picroside-I, Picroside-II and Kutkoside. Callus was induced on Murashige and Skoog basal medium supplemented with cytokinin (Kn or BA) and auxin (2,4-D/IBA/NAA) using leaf, petiole and stem explants.   • A method for micropropagation was developed for Dendrocalamus asper, an economically and environmentally important bamboo. Disinfected seeds were cultured in flasks containing 20 ml of Murashige and Skoog’s medium (MS) supplemented with BA (1.0-7.0 mg l-1 ) or kinetin (1.0-7.0 mg l-1 ). Multiple shoots (6.53) were formed on MS medium supplemented with 3.0 mg l-1 BA and reached 1.49 cm in length. Continuous shoot proliferation was achieved on a MS medium supplemented with BA (1.0-7.0 mg l -1 ). The multiplication rate of 3.30 fold was achieved on MS medium supplemented with 3.0 mg l-1 BA.

  • The present study develops a protocol for rapid in vitro micropropagation of a critically endangered and floriculturally most important epiphytic orchid, Dendrobium primulinum Lindl. through the culture of small shoot tip explants (0.3 to 0.5mm) derived from in vitro grown seedlings. The shoot tip explants cultured on solidified Murashige and Skoog (MS) basal medium and MS medium alone or supplemented with combination of various concentrations of growth regulators; α-naphthalene acetic acid (NAA) and 6 benzylaminopurine (BAP), produced shoots and multiple shoots.
