In vitro effect of culture filtrates

In in vitro evaluation of cultural extract of Chaetomium globosum against Fusarium, Sclerotium and Rhizoctonia, it was observed that inhibition of pathogen was increased with increase in concentration of metabolite indicating the antifungal properties against pathogen. Chaetomium globosum showed maximum antagonistic effect to Fusarium (51.50%) at 100% concentration. Significantly minimum inhibition zone (20%) was observed at 25% for Fusarium spp. Effect of culture filtrate did not show any inhibition effect against soil borne pathogens Sclerotium and Rhizoctonia.

Antagonistic potentiality of four isolates of Trichoderma species viz. Trichoderma viride isolate A, Trichoderma viride isolate M, Trichoderma harzianum isolate C and Trichoderma harzianum isolate L against Beauveria bassiana causing white muscardine disease of Silk worm, Bombyx mori L, was studied invitro by studying the mycelial interaction between them as well as by the effect of culture filtrate of biocontrol agents on sporulation and spore germination of pathogen.

Five rhizospheric bacterial cultures and cell free culture filtrates (CFs) (one strain of Gluconacetobacter diazotrophicus (35-47), two strains of Azotobacter chroococcum viz. AVK-51, HT-54: two strains of Bacillus spp. viz., RKB-91, RKB-65) were evaluated for larval mortality and egg hatching against rice root-knot nematodes (Meloidogyne gramincola) at CCS HAU Hisar.

Four fungal bioagents viz., Trichoderma viride, T. harzianum, Pochonia chlamydosporia and Purpureocillium lilacinum were screened for their efficacy against Meloidogyne incognita under in-vitro conditions through egg parasitism, egg hatch inhibition and second-stage juveniles mortality test. P. chlamydosporia showed highest egg parasitism. In respect of egg hatch inhibition and juvenile mortality, the culture filtrates of these fungal bioagents were tested and found to be effective in inhibition of egg hatch and mortality of juveniles of M.

Silver nano particles were synthesized using two potential isolates of Trichoderma, Psudomonas fluorescens and one isolate of pathogen Rhizoctonia solani cell free culture filtrates incubated at different days 0, 5, 10 and 15. The present investigation revealed that bionano preparations irrespective of the isolate source used, i.e., Trichoderma, P. fluorescens or R. solani were found better in decreasing the sheath blight incidence in vitro though variation existed in terms of quantum of disease reduction.

The aim of the present experiment was to observe the effect of extracellular metabolites of the biocontrol agent Trichoderma pseudokoningii on the radial growth of four selected pathogens viz. Fusarium oxysporum, Colletotrichum capsici, Rhizoctonia solani and Sclerotinia sclerotiorum in in vitro conditions.

Non-pathogenic Fusarium isolates were isolated and five isolates were selected for their biocontrol activity against Fusarium wilt of chrysanthemum. Different methods of in vitro tests were conducted, under the dual plate method the isolate UASB NPF-III and UASB NPF-I inhibited the pathogen by 64.17 % and 60.44 % respectively at seventh day of incubation. The cell free culture filtrates of non pathogenic Fusarium isolates UASB NPF-III and UASB NPF-I were found effective in controlling the pathogen by 26.36 % and 24.34 % at seventh day of inoculation.

We succeeded in large volume artificial cultivation of Onygena corvina. The appearance (shape, color, and size) of fruiting bodies of cultured bacteria was almost the same as that of wild one. We screened their metabolites for antiproliferative activities against nine human cancer cells in vitro. Culture filtrates (metabolite-containing media) of O. corvina were lyophilized and resuspended in phosphate buffer to investigate them in vitro. As a result, these metabolites effectively inhibited the growth of some cancer cells in a concentration-dependent manner.

Penicillium expansum is a post-harvest pathogen of apples which can produce the hazardous mycotoxin patulin. The yeast Cryptococcus laurentii (LS28) is a biocontrol agent able to colonize highly oxidative environments such as wounds in apples. In this study culture filtrates of the basidiomycete Lentinula edodes (LF23) were used to enhance the biocontrol activity of LS28. In vitro L. edodes culture filtrates improved the growth of C. laurentii and the activity of its catalase, superoxide dismutase and glutathione peroxidase, which play a key role in oxidant scavenging.
