Ebook "Retinal gene therapy: Methods and protocols" describes a spectrum of methods and protocols that can be used for the bench-to-bedside development and evaluation of retinal gene therapy. Methods for the successful delivery of these gene therapy vector systems to the retina are examined, as well as assays to test the efficacy in vitro in cell cultures, for gene augmentation and gene editing in vivo on rodents, pigs, and monkey retinas, and on human retinal explants as well as in human clinical studies.

The species belonging to the Lamiaceae family have been useful tools for many years as a folk remedy. In the present study, a microemulsion system was developed with extract of Scutellaria salviifolia Benth (endemic), Sideritis libanotica Labill. subsp. linearis (Bentham) Bornm (endemic), and Ziziphora clinopodioides Lam. Both and related microemulsion formulations were also investigated for their potential biological activities.

The aim of the present study was to synthesize and characterize benzothiazine derivatives prepared by using 2-aminothiophenol and saccharine and in vitro screen their enzyme inhibition and antioxidant potential. Nine different derivatives were prepared and their structures were confirmed by spectral studies (EIMS, 1H and 13 C NMR). Enzyme inhibition potential of the synthesized compounds was evaluated by Ellman’s method, while antioxidant activity was determined by DPPH and FRAP assays.

A new, less expensive, solvent-free procedure was developed for the synthesis of some new derivatives of various fused heterocyclic ring systems, namely azolopyridazine, azolotriazine, azinotriazine, thienopyridine, and pyrazolopyridine. The structures of the products prepared were established by their spectral data and elemental analyses. Eight compounds were evaluated for their in vitro antimicrobial activity. Some of the tested compounds exhibited moderate to significant antibacterial and antifungal activities.

This study aimed to estimate the effect of three concentrations (75 µg/mL, 150 µg/mL and 300 µg/mL) of the European raspberry (Rubus iadeus) on the selected quality parameters of bovine spermatozoa after 0h, 2h, and 24h of in vitro culture. Sperm motility was evaluated using the Computer-assisted sperm analysis (CASA) system. The mitochondrial activity was detected by the metabolic (MTT) assay.

: In this study, magnetic O-carboxymethyl chitosan (MOCC) nanocomposite was synthesized and characterized as a drug delivery system for loading the anticancer drug irinotecan (CPT-11). To increase the drug loading capacity, MOCC was synthesized by linking the carboxyl group functionally to chitosan. Also, several critical factors such as concentration, the dose of MOCC, and contact time for optimum drug loading condition were investigated. The loading capacity of CPT-11 onto MOCC was calculated as 5.6 mg/g, and the loaded drug concentration was calculated as 0.04787 mM at pH value of 5.

. In this study, the VEGF121 gene was cloned and expressed in a prokaryotic expression system (Escherichia coli). The recombinant VEGF121 was encapsulated with PLA microparticles and studied in vitro and ex ovo for the sustained release mechanism. The PLA-VEGF microparticles and the recombinant VEGF121 were explored for their bioactivity in human umbilical vein endothelial cells (HUVEC).

In vitro evaluation of three contact and five systemic and two combination was carried out under laboratory condition in 2018-19 at Department of Plant Pathology, College of Horticulture, Mysuru by using Potato Dextrose Agar medium (PDA) as basal medium and six biocontrol agents were evaluated through dual culture technique. The result revealed that, there was significant difference among the fungicides in inhibiting the radial growth of the A. solani.

Six fungicides viz., captan 50 WP, copper oxychloride 50 WP, dodine 65 WP, mancozeb75 WP, propineb70 WP and zineb 80 WP were evaluated against rice sheath blight (Thanatephorus cucumeris). The in-vitro evaluation of these fungicides through poisoned food technique at five different concentrations viz., 50, 100, 200, 500 and 1000 µg a.i ml-1 indicated that mancozeb proved the most effective exhibiting mean mycelial growth inhibition of 95.26 per cent followed by captan and dodine with 93.80 and 93.66 per cent, respectively. Copper oxychloride proved least effective and resulted in only 44.

Twelve fungicides including systemic and non-systemic, five bioagents and eight plant extracts were evaluated in vitro against Exserohilum turcicum, the causal agent of Turcicum leaf blight of maize. All the test fungicides, bioagents and plant extracts showed fungistatic action and significantly inhibited mycelial growth of the test pathogen over untreated control.

An in vitro bioassay was undertaken to record the compatibility of local isolate of Trichoderma harzianum with fungicides used in coorg mandarin-pepper-coffee plantations. Eight non-systemic, 10 systemic and 9 combi products fungicides were evaluated against Trichoderma for radial growth inhibition on PDA medium using poisoned food technique. Contact fungicides at selected concentration were found to be safer than systemic and combi products except Chlorothalonil.

Chickpea (Cicer arietinum L.) is one of the major legume pulse crops and it is majorly grown in India and other semi-arid regions of the world. Dry root rot of chickpea is the most destructive disease of chickpea. Therefore, in present studies fungicides (six systemic, seven non systemic and six ready mix fungicides) were tested in vitro at three different concentrations by poisoned food technique for evaluating their efficacy against M. phaseolina.

Genetic uniformity of in vitro raised plants is a prerequisite for production of quality planting material of banana. Embryogenic cultures were induced from immature male flower buds of two important cultivars of banana, ‘Rasthali’ (Silk-Musa AAB) and ‘Grand Naine’ (Cavendish-Musa AAA). Cell suspensions were established and regenerated plants were evaluated for their genetic stability using 10 inter simple sequence repeat markers.

Foot rot of tomato is an important disease which causes severe loss in tomato production. The present study was carried out to evaluate five bioagents and six fungicides against Sclerotium rolfsii. Among the five antagonists tested the maximum inhibition was observed in Trichoderma virens (67%) and contrarily, no inhibition was observed in Bacillus subtilis. Six fungicides viz., carbendazim, captan, tebuconozole, copper oxychloride, hexaconozole and Tebuconazole +Trifloxytrobin at different concentrations (50, 100, 150, 200, 250 and 500 ppm), were tested.

In vitro evaluation of some fungicides (systemic and non-systemic) against twenty different Indian isolates of Sclerotium rolfsii were tested on potato dextrose agar medium by using poisoned-food technique and per cent growth inhibition was recorded at ranging from 125, 250 and 500 ppm for systemic; and 500, 1000 and 2000 ppm for non-systemic, respectively. For fungicides evaluation, the isolates were grouped on the basis of growth rates on Petri plates with 90 mm diameter as fast (5-6 days), medium (8-10 days) and slow growing isolates (12-15 days) respectively.
