  • This study investigated the effects of different PGRs on in vitro plant regeneration of Anthurium scherzerianum Schott and acclimatization in the ex vitro stage. Depending on the objective of each experiment, the in vitro shoots were induced from callus cultured on a modified Murashige and Skoog (MS) medium containing cytokinins and auxins.

  • The shoots in vitro were transferred to half-strength MS supplemented with different concentrations of IAA, IBA and NAA to investigate root formation. The best rooting occurred at 0,5 mg/L NAA (7.91 roots/shoot; root length of 4.01 cm; 98.51% root formation). The plantlets with uniform growth were planted on different substrate: Eco clean soil, Coconut fiber, Fern fiber, 50% Rice husk in combination with 50% Eco clean soil for research the most suitable substrate.

  • Rhizoctonia root rot of okra caused by Rhizcotonia solani is an important disease which is known to cause heavy loss in Rajasthan as well as in India. Among physiological parameters effect of temperature, effect of pH and effect of relative humidity were study in vitro. The fungus grew at all the temperature level ranged from 15ºC to 35ºC. Maximum mycelial growth of the fungus (90mm) was observed at 30ºC. In different pH levels that maximum dry mycelium weight (630gm) of Rhizoctonia solani was observed at 6.5 pH level.

  • Root rot of soybean is an important disease of soybean inflicting heavy losses in crop. The present investigation was carried out to test the efficacy of agrochemicals and fertilizers in vitro. In bioassay test of non systemic fungicides, Zineb 75 WP gave cent per cent inhibition of growth and sclerotial formation at minimum concentration of 500 ppm followed by copper hydroxide 77WP (53.63%).

  • The effects of different culture media and temperature on mycelial growth and sclerotial formation of Macrophomina phaseolina (Tassi) Goid inciting agent of castor root rot were studied in vitro. Eight different solid and liquid media were tested for their effect on mycelial growth and sclerotial formation of test fungus. Among the solid media, potato dextrose agar (90.00 mm) and Richard's agar (87.71 cm) were the best for fungus growth as well as for sclerotial formation followed by Elliott's medium (80.08 cm).

  • Papaya is one of the few important fruits which flowers and yield throughout the year. Though a polygamous species with three basic types viz., staminate, pistillate and bisexual, its cultivation with seed propagation is hindered by problems due to vide variability in sex expression and fruit characters. In such point of view, propagation through asexual method could be a better alternative. The micropropagation will substitute method of propagation by cutting or grafting would be of great boon to papaya industry. The procedure for in vitro propagation of papaya (Carica papaya) cv.

  • Arbuscular Mycorrhizal Fungi benefits plants by improving the uptake of phosphate and other nutrients from soil and also increases the disease tolerance to the host plant. The fungal pathogens cause huge loss to vegetable crops when infected. In order to overcome this problem, the influence of AMF in the control of fungal plant pathogens is studied. AMF, Glomus sp. was isolated from the rhizosphere soil of tomato plants. The wilt pathogen, Fusarium oxysporum f.sp. lycopersici was cultured from the infected roots of tomato plants.

  • The results showed that protocorms were formed from seeds culture on the ½ MS medium supplemented with 10% potatoes extract within 8 weeks. Protocorms which were cultured on the ½ MS medium containing 30 g/l sucrose, 0.5 g/l AC, 7 g/l agar, 0.1 mg/l NAA and 1 mg/l BA were optimal for PLB formation (8.67 PLBs/sample) after 8 weeks culture. Protocorms converted into normal plants with well-developed shoots and roots on the ½ MS medium supplemented with 20 g/l sucrose, 10% potatoes extract, 0.5 g/l AC, and 7 g/l agar after about 90 days.

  • Chilli is grown in many states in India as a valuable trade crop. Chilli accounts for 20-30% of total Indian spices exports valuing approx Rs. 400-500 cores. Nine cultural media Potato dextrose agar, Host extract agar (Chilli), Oat meal agar, Carrot agar, Plane agar, Richards agar, Czapeks agar, Elliots agar, Saboroud’s agar were analyzed for in vitro mycelial growth of Sclerotium rolfsii. The cultural characteristics indicated that the excellent growth and sclerotia formation were observed on different semi-synthetic media viz.

  • This study was carried out to assess the propagation of Tacca leontopetaloides (Taccaceae) in Vitro using seed embryos. The seeds were cultured on Murashige & Skoog basal media augmented with various concentrations of single or combined growth hormones to obtain a suitable protocol for plantlet regeneration. Seed embryo germination of 57% was obtained, as against 20% for germination under conventional sowing. The best shoot proliferation was obtained using media fortified with 0.1 mg/L 6-benzyl amino purine and 0.
