In this study, human induced pluripotent stem cells (hiPSCs) were successfully differentiated into cardiomyocytes and subsequently used to generate spheroids via the hanging drop method. We comprehensively characterized the formation, fusion, and beating capability of the generated spheroids.

This fraction and the ethanol extract from green tea were also able to induce the expression of hyperpolarizationactivated cyclic nucleotide-gated channel 4 (HCN4), the important factor for automatic beating of SAN, but insignificantly (P > 0.05). In conclusion, neonatal cardiomyocytes were isolated and suitable for use as a model for screening of pacemaker cell transduction effects of potential candidates. From this study, the garlic ethyl acetate fraction presented as the most promising nominee for further neonatal cardiomyocyte transdifferentiating effects.

Induced Pluripotent stem cells (iPSC) technology is involved in advancing the field of disease modelling and cell transplantation. Stem cell can differentiate under appropriate in vitro and in vivo conditions into different cell types. Under this study, we analysed the establishment of in vitro expression of GATA4 gene in cardiac differentiated canine iPSC. In vitro differentiation of canine iPSCs via embryoid bodies (EBs) was produced by ‘Hanging Drop’ method. EB’s were differentiated by using cardiac differentiation media.
