Our immune system has its self-regulation mechanisms. Leukocytes can regulate their functions via control of the gene expression and/or the secretion of cytokines. In the review "Cancer immunotherapy: Immune-checkpoint blockades", we resumed the way T-cells are regulated between active and inactive states. The discovery of two receptors for the transduction of inhibitory signals, the cytotoxic T-cell receptor and the programmed cell death receptor, is presented in the second part.

Inflammasomes are supramolecular protein complexes implicated in the detection of pathogens or danger-associated molecules and are responsible for mounting the first line of innate immune response to counteract these signals and restore tissue homeostasis. Among different inflammasomes identified so far, NLRP3 is of main interest since mutations in Nlrp3 gene are associated with autoinflammatory diseases such as Muckle–Wells syndrome, neonatal onset multisystem inflammatory disease, and familial cold urticaria/autoinflammatory syndrome.

Disruption of the transforming growth factor-beta (TGF-β) signaling pathway is observed in many cancers, including cervical cancer, resulting in TGF-β resistance. While normal human keratinocytes (HKc) and human papillomavirus type 16-immortalized HKc (HKc/HPV16) are sensitive to the growth inhibitory effects of TGF-β, HKc/HPV16 develop resistance to TGF-β1 as they progress in vitro to a differentiation resistant phenotype (HKc/DR).
