VEGF is one of the key drivers of physiological or pathological angiogenesis hence several VEGF inhibitors are in different stages of clinical development. To further dissect the role of VEGF in different stages of tumor progression in lung tumors, we utilized KrasG12D-LSL GEMMs (genetically engineered mouse models).

Certain amino acids, like glutamine and leucine, induce an anabolic response in liver. They activate p70 ribosomal protein S6 kinase (p70S6K) and acetyl-CoA carboxylase (ACC) involved in protein and fatty acids synthesis, respectively. In contrast, the AMP-activated protein kinase (AMPK), which senses the energy state of the cell and becomes activated under metabolic stress, inactivates by phosphorylation key enzymes in biosynthetic pathways thereby conserving ATP.

Aberrant regulation of protein kinases impairs normal cellular functioning and may lead to disease. The protein kinase involved in the regulation of the dynamics of the actin cytoskeleton, Rho-kinase (ROCK), phosphory-lates various substrates (e.g. myosin light chain, myosin phosphatase), caus-ing the formation of actin fibers and tension inside cells.

We report the development of a high-level bacterial expression system for the Alzheimer’s disease-associated amyloid b-peptide (Ab), together with a scaleable and inexpensive purification procedure. Ab(1–40) and Ab(1–42) coding sequences together with added ATG codons were cloned directly into a Pet vector to facilitate production of Met-Ab(1–40) and Met-Ab(1– 42), referred to as Ab(L1–40) and Ab(L1–42), respectively.
