Liposomal nanoparticles

Mesoporous silica nanoparticles (MSNs) have been used as an anticancer drug delivery system with high safety and entrapment capacity thanks to their large internal space for drug accommodation, durable structure, and good biocompatibility. However, the treatment efficiency of the bare MSNs is limited due to its drug leakage and burst release. In this study, a phospholipid bilayer was covered on the MSNs surface (MSN@Lip) as a liposomal cap that not only reduced drug leakage but also improved the stability of the colloidal system.

This study aims to establish promising and simple MNP-based protocol for gene delivery initially. Specifically, we aim to evaluate whether sodium chloride (NaCl) could enhance gene transfer with naked plasmid DNA without the need to combine liposomes or polymersomes.

PEP02 (also known as MM-398, nal-IRI) is a novel nanoparticle formulation of irinotecan encapsulated in liposomes. The aims of this study were to investigate the dose-limiting toxicity (DLT), maximum tolerated dose (MTD) and pharmacokinetics (PK) of PEP02 in combination with 5-FU and LV, in patients with advanced refractory solid tumors.

Quercetin, a pigment (flavonoid) found in many plants and foods, has good effects on protecting liver function but poor solubility and bioavailability in vivo. A drug delivery system can improve the accumulation and bioavailability of quercetin in liver. In this study, we used liposomal nanoparticles to entrap quercetin and evaluated its protective and therapeutic effects on drug-induced liver injury in rats.

Rosuvastatin (RSV) is a poorly water-soluble drug that has an absolute oral bioavailability of only 20%. The aim of this work was to prepare a positively charged chitosan coated flexible lipid-based vesicles (chitosomes) and compare their characteristics to the corresponding negatively charged flexible liposomal nanoparticles (NPs) in order to develop new RSV nanocarrier systems.

Nonspecific distribution and uncontrollable release of drugs in conventional drug delivery systems (CDDSs) have led to the development of smart nanocarrier-based drug delivery systems, which are also known as Smart Drug Delivery Systems (SDDSs). SDDSs can deliver drugs to the target sites with reduced dosage frequency and in a spatially controlled manner to mitigate the side effects experienced in CDDSs. Chemotherapy is widely used to treat cancer, which is the second leading cause of death worldwide.

With parallel breakthroughs occurring in molecular biology and nanoscience/technology, the newly recognized research thrust on “nanomedicine” is expected to have a revolutionary impact on the future of healthcare. To advance nanotechnology research for cancer prevention, diagnosis, and treatment, the United States National Cancer Institute (NCI) established the Alliance for Nanotechnology in Cancer in September 2004 and has pledged $144.3 million in the next five years (for details, visit http://nano.cancer.gov).
