In a previous study, a gene GL0694641 coding for endoglucanase containing 3 domains GH5- CBM72-CBM72 was exploited from metagenomic DNA data of bacteria in Vietnamese goats’ rumen. The gene (eg5) encoding the mature enzyme (without signal peptide coding sequence) was optimized codons, artificially synthesized, and inserted into the pET22b(+) vector at NcoI and XhoI to generate expression vector pET22-eg5 for expression of the gene in Eschrichia coli.

Proteases represent one of the most abundant classes of enzymes in eukaryotes and are known to play key roles in many biological processes in plants. However, little is known about their functions in fruit ripening and disease resistance, which are unique to flowering plants and required for seed maturation and dispersal.

Obligate sulfur oxidizing chemolithoauthotrophic strains of Hydrogenovibrio crunogenus have been isolated from multiple hydrothermal vent associated habitats. However, a hydrogenase gene cluster (encoding the hydrogen converting enzyme and its maturation/assembly machinery) detected on the first sequenced H. crunogenus strain (XCL-2) suggested that hydrogen conversion may also play a role in this organism.

Weather change in high-altitude areas subjects mature tobacco (Nicotiana tabacum L.) to cold stress, which damages tobacco leaf yield and quality. A brupt diurnal temperature differences (the daily temperature dropping more than 20 °C) along with rainfall in tobacco-growing areas at an altitude above 2450 m, caused cold stress to field-grown tobacco.

Thus far, a handful of genes have been shown to be related to the wing maturation process in insects. A novel heme peroxidase enzyme known as curly suppressor (Cysu)(formerly CG5873), have been characterized in this report because it is involved in wing morphogenesis.

Dicer is necessary for the process of mature microRNA (miRNA) formation because the Dicer enzyme cleaves pre-miRNA correctly to generate miRNA with correct seed regions. Nonetheless, the mechanism underlying the selection of a Dicer cleavage site is still not fully understood.

Sialyltransferase I (ST6Gal-I) is an enzyme involved in tumor metastasis that processes sialic acid precursors into their mature form, enabling them to regulate gene expression. However, the effect of ST6Gal-I on the biological behavior of cancer cells remain unclear.

Specific activity of two hydrolytic enzymes viz., cellulase and polygalactouranase was assayed in the shattering and non-shattering zones of pod wall and lignin content in pod wall of soybean varieties at four harvesting stages viz., at physiological maturity, 5 DAPM, 10 DAPM and 15 DAPM. The cellulase enzyme activities was lowest at non-shattering zone of pod wall of the susceptible varieties as compare to the resistant varieties but highest at shattering zone of pod wall of the susceptible varieties indicating its role in pod shattering.

Soil health indicators such as soil physico-chemical properties, total bacterial count, microbial enzyme activities were monitored at different crop growth stages (such as before ploughing, after transplantation (0 DAT), max tillering (45 DAT), flowering (60 DAT) and maturity (90 DAT) of rice variety Pusa Basmati 1. The treatment comprises green manure Sesbania rostrata and inorganic fertilizers for organic and conventional respectively. Moreover, field was cultivated with rice variety Pusa Basmati I.

Soil carbon dynamics in oil palm - Cocoa cropping system in comparison to that of oil palm sole crop
An investigation was conducted in oil palm sole crop and oil palm + cocoa cropping systems to study microbial diversity along with enzyme activities and understand soil carbon dynamics in oil palm based cropping system. Ten mature oil palm plantations (five each in oil palm sole crop and oil palm + cocoa) planted with Tenera hybrids with a spacing of 9 X 9 X 9 m in a triangular system were selected for the study. Cocoa var. Forastero is cultivated as an intercrop in oil palm plantations at 4.5 X 4.5 m spacing.

The field experiment was conducted with maize during kharif on a sandy loam soil at College Farm, College of Agriculture, Rajendranagar, Hyderabad. The experiment was laid out in Randomized Block Design with 13 treatments, each replicated thrice. The urease and dehydrogenase activites in soil were assayed and the results showed an increasing trend in enzymatic activities with the age of the crop and exhibited highest activity at tasseling stage and thereafter the activity decreased towards maturity and harvest.

Honey is one of the oldest and best loved sweetening agents for foods and over the centuries, it has still retained a “natural image”. The raw materials for the production of „Floral honey‟ are nectar, a dilute solution of sugars found in the nectarines of the flowering plants.

Changes in antioxidative enzymes of young and mature leaves of tomato seedlings under drought stress
The effects of exogenous abscisic acid (ABA) on antioxidant enzymes of young and mature leaves of 4-week old Lycopersicon esculentum cv. Ailsa Craig and ABA-deficient mutant, notabilis, were investigated under drought stress. Although ABA induced increases in ascorbate peroxidase (APX), glutathione reductase (GR) and catalase (CAT) activities in drought-stressed young leaves of notabilis, CAT activities remained unchanged in both ABA-treated young and mature leaves of Ailsa Craig under drought.

Two-way anova analyses showed that changes of TPC and AC of cocoa beans were dependent on maturity stages, fermentation methods and fermentation duration. As a conclusion, fermentation of cocoa beans harvested at the maturity stage two and three was found to be optimum for the high levels of AC and TPC attainment; fermented beans with commercial enzyme could be utilized to reduce labor cost by shortening the fermentation duration.

Arecombinant streptococcalC5apeptidasewas expressed in Escherichia coliand its catalytic properties and thermal sta-bility were subjected to examination. It was shown that the NH2 -terminal region ofC5a peptidase (Asn32–Asp79/ Lys90) forms the pro-sequence segment. Upon maturation the propeptide is hydrolyzed either via an autocatalytic intramolecular cleavage or by exogenous protease strepto-pain. At pH 7.4 the enzyme exhibited maximum activity in the narrow range oftemperatures between 40 and 43C....

Genes homologous to hydrogenase accessory genes are scattered over the whole genome in the cyanobacteriumSynechocystissp. PCC 6803. Deletion and insertion mutants of hypA1 (slr1675), hypB1 (sll1432), hypC, hypD, hypE and hypF were constructed and showed no hydrogenase activity. Involvement of the respective genes in maturation of the enzyme was con-firmed by complementation. Deletion of the additional homologueshypA2 (sll1078) and hypB2(sll1079) had no effect on hydrogenase activity. Thus, hypA1andhypB1are specific for hydrogenase maturation....

Mature, active mammalian histidine decarboxylase is a di-meric enzyme of carboxy-truncatedmonomers (53 kDa). By using a biocomputational approach, we have generateda three-dimensional model of a recombinant 1/512 fragment of the rat enzyme, which shows kinetic constants similar to those of the mature enzyme purified from rodent tissues. This model, together with previous spectroscopic data, al-lowed us to postulate that the occupation of the catalytic center by the natural substrate, or by substrate-analogs, would induce remarkable changes in the conformation of the intact holoenzyme....
