Dendrimeric peptides selective for microbial surfaces have been developed to achieve broad antimicrobial activity and low hemolytic activity to human erythrocytes. The dendrimeric core is an asymmetric lysine branching tethered with two to eight copies of a tetrapeptide (R4) or an octapeptide (R8). The R4 tetrapeptide (RLYR) contains a putative microbial surface recognition BHHB motif (B ¼ basic, H ¼ hydrophobic amino acid) found in protegrins and tachyplesins whereas the octapeptide R8 (RLYRKVYG) consists of an R4 and a degenerated R4 repeat....

Here we show, for the ®rst time, thein vitro formation of ®lamentous aggregates of phosphorylated tau protein in SH-SY5Y human neuroblastoma cells. The formation of suchaberrant aggregates, similar to thoseoccurringinvivoin Alzheimer's disease and other tauopathies, requires okadaic acid, a phosphatase inhibitor, to increase the level of phos-phorylated tau, and hydroxynonenal, a product of oxidative stress that selectively adducts and modi®es phosphorylated tau.

There is much evidence to indicate that cholesterol forms lateral membranemicrodomains (rafts), and to suggest their important role in cellular signaling. However, no probe has been produced to analyze cholesterol behavior, especially cholesterol movement in rafts, in real time. To obtain a potent tool for analyzing cholesterol dynamics in rafts, we prepared and characterized several truncated fragments of h-toxin (perfringolysin O), a cholesterol-binding cytolysin, whose chemicallymodified formhas been recently shown to bind selectively to rafts. ...

Site-directedmutagenesis was used to investigate the control of 2-oxoacid cosubstrate selectivity by deacetoxycephalo-sporin C synthase. The wild-type enzyme has a requirement for 2-oxoglutarate and cannot efficiently use hydrophobic 2-oxoacids (e.g. 2-oxohexanoic acid, 2-oxo-4-methyl-penta-noic acid) as the cosubstrate. The followingmutant enzymes were produced: R258A, R258L, R258F, R258H and R258K. All of the mutants have broadened cosubstrate selectivity and were able to utilize hydrophobic 2-oxoacids.

A library of random peptide sequences was used to select peptides that inhibit an anti-idiotypic catalytic Ig, immuno-globulin (IgG) 9G4H9, withab-lactamase-like activity. This library displays cyclic heptapeptides on the surface of bac-teriophagesand representsacollectionofupto4.5·10 9 peptides. The first selection step aimed at enriching the lib-rary in species that bind to the whole Ig molecule. The sec-ond stepwas todiscriminate peptides that bind topart of the molecule other than the active site. Selected peptides were then screened by surface plasmon resonance analysis.

In aqueous media, muscle pyruvate kinase is highly selective for K + over Na + . We now studied the selec-tivity of pyruvate kinase in water/dimethylsulfoxide mix-tures by measuring the activation and inhibition constants of K + and Na + , i.e. their binding to the monovalent and divalent cation binding sites of pyruvate kinase, respect-ively [Melchoir J.B. (1965) Biochemistry4, 1518–1525]. In 40% dimethylsulfoxide theK0.5 appfor K + and Na + were 190 and 64-fold lower than in water.Kiappfor K + and Na +decreased 116 and 135-fold between 20 and 40% dimethylsulfoxide. The ratios of Kiapp/K0.

The dimeric structure of seminal ribonuclease (BS-RNase) is maintained by noncovalent interactions and by two intersubunit disulfide bridges.Another unusual feature of this enzyme is its antitumour action, consisting in a cyto-toxic activity selective for malignant cells. This cytotoxic action is exerted when the protein reaches the cytosol of the affected cells, where it degrades ribosomal RNA, thus blocking protein synthesis and leading cells to death.

The Hsp90 molecular chaperone catalyses the final activa-tion step of many of themost important regulatory proteins of eukaryotic cells. The antibiotics geldanamycin and rad-icicol act as highly selective inhibitors of in vivo Hsp90 function through their ability to bindwithin the ADP/ATP binding pocket of the chaperone. Drugs based on these compounds are now being developed as anticancer agents, their administration having the potential to inactivate sim-ultaneously several of the targets critical for counteracting multistep carcinogenesis. ...

Heteronuclear NMR relaxation measurements and hydro-gen exchange data have been used to characterize protein dynamics in the presence or absence of stabilizing solutes from hyperthermophiles. Rubredoxin from Desulfovibrio gigaswas selected as a model protein and the effect of diglycerol phosphate on its dynamic behaviour was studied. The presence of 100 mMdiglycerol phosphate induces a fourfold increase in the half-life for thermal denaturation ofD.

Cancer cells develop strong anti-apoptotic signaling pathways and therefore escape many therapeutic regimens. Recognizing this feature of cancer cells, we have focused on two approaches: dis-arming the cancer cell from its anti-apoptotic weaponry [1, 2] and applying strategies aimed at enhancing pro-apoptotic signa-ling pathways selectively in the cancer cell [3]. The first goal has been achieved by developing highly selective Aktstatins [1, 2] that inhibit PKB 100 times better than PKA or PKC.

Eukaryotic TGs evolved from the papains, but utilizing a Trp/ Cys/His/Asp catalytic tetrad instead of Gln/Cys/His/Asp for acy-lation-deacylation. Because saturable binding of specific amine substrates, aminolysis is preferred over hydrolysis in deacylation. Transamidation leads to protein remodelling of the gamma amides of select Gln residues by crosslinking to epsilon amines of select Lys’s or by incorporation of small primary amines. Such post-translational modifications have important biological conse-quences. ...

We have used the circular dichroismand infrared spectra of a specially designed 50 protein database [Oberg, K.A., Ruysschaert, J.M. & Goormaghtigh, E. (2003)Protein Sci. 12, 2015–2031] in order to optimize the accuracy of spectroscopic protein secondary structure determination using multivariate statisticalanalysis methods. The results demonstrate that when the proteins are carefully selected for the diversity in their structure, no smaller subset of the database contains the necessary information to describe the entire set....

Cellular functions are ultimately linked to metabolic fluxes brought about by thousands of chemical reactions and transport processes. The synthesisof theunderlying enzymes and membrane transporters causes the cell a certaineffort of energyandexternal resources.Considering that those cells should have had a selection advantage during natural evo-lution that enabled them to fulfil vital functions (such as growth, defence against toxic compounds, repair of DNA alterations, etc.)

Three protein factors IF1, IF2 and IF3 are involved in the initiation of translation in prokaryotes. No clear function has been assigned to the smallest of these three factors, IF1. Therefore, to investigate the role of this protein in the initi-ation process inEscherichia coliwe have mutated the cor-responding gene infA. Because IF1 is essential for cell viability and no mutant selection has so far been described, the infA gene in a plasmid was mutated by site-directed mutagenesis in a strain with a chromosomalinfA + gene, followedbydeletionof thisinfA + gene.

For 30 years, the field of medical device technology has struggled to identify the specifications for optimal biocompatibility. For the vast majority of devices, clinical experience has eventually confirmed that the best performance is achieved when biocom-patibility is equated with biological safety that is materials are selected on the basis of maximizing bioinertness and minimizing toxicity and irritation.
